PYRRHOTITE has long been considered a gangue mineral to be eliminated as tailing in the treatment of various sulphide ores. However, in recent years the world-wide lack of sulphur resources has called attention to this mineral as a potential source of both sulphur and iron. Its importance as an economic mineral, however, has not been particularly emphasized. For this reason very little is known about its response to flotation, except that it can be depressed easily in alkaline circuit, by long aeration,1,2 addition of oxidizing agents3 or by starch.4 The object of this work was to study the floatability of pyrrhotite. This includes the effect of oxidation by aeration, of copper activation, and of change in pH.

Multiple operating points for mine fans are a problem that fan and mining engineers must constantly be aware of in mine/fan systems. In a single-fan system, unstable conditions can be avoided by restricting fan operation to the "normal operating range." When mine fans are operated in parallel, the situation becomes a little more complicated. Multiple operating points are more often encountered and the fan user must be aware of these multiple points to have a suitable set of operating conditions. However, for fans in parallel, the engineer can plot the combined characteristic in a relatively straightforward manner and thus can avoid conditions that would cause instability in the mine/fan system.

The analysis of Kikuchi pattersns of exct ovientalions from single cryslals and paired Kikuchi lines from single and overlapping crystals is shown to be useful and quanlitalve and is applied to Phase transfovnzcitions including ordering, spinodals, mavten-silic, and nuclealion and growth pyocesses. 112 pinciple, the analysis of exacl orientations enables the crystal system and the Bravais lattice of a crystal to he determined. The advantages of the davk-field imaging technigure for detecting vevy small precipitates are also described. ALTHOUGH Kikuchi electron-diffraction patterns were first observed nearly 40 years ago,' little systematic application seems to have been made of them, until fairly recently during electron microscopy, when their usefulness in contrast experimen and for determining exact orientations8'9 has been pointed out. With the availability of gonio-metric specimen tilting stages it has now become possible to make much wider use of diffraction patterns, particularly the Kikuchi pattern, which is the main subject of this paper. The treatment presented here is not exhaustive but it is hoped that it will stimulate more use of Kikuchi patterns during electron-microscopy investigations. The Kikuchi pattern is formed as a result of Bragg diffraction of the inelastically scattered electrons produced during the interaction of the beam and thick specimens, The important feature of these patterns is that they give an accurate representation of the symmetry of the crystal being investigated, so that it is possible to identify crystal systems and even the Bravais lattice. This means that new structures, e.g., formed in phase transformations, may be identified during normal electron microscopy, so that Kikuchi-pattern analyses considerably extend the uses of the electron microscope. Recent work has also shown that dark-field images are more informative than bright-field images, particularly, for observing small precipitates. The second part of this paper discusses some applications and advantages of dark-field imaging in studies of two-phase systems. 1) DIFFRACTION PATTERNS 1.1) Spot Patterns. Electron-diffraction spot patterns have their limitations because of the importance of the form factor on the intensities and shape of the reciprocal lattice points. Because of the extension of these points into relrods, reflections are possible over a large angular range (+5 deg) and the patterns from thin regions can be complicated because second-layer relrods intersect the reflecting sphere. Spot patterns can thus give only an approximate idea of the crystallography unless the foil is tilted into exact orientation. In this case the spot pattern is symmetrical, with equal numbers of spots on the positive and negative zone directions about the origin. Such cases are necessary for structural analyses and have been used recently to determine the crystal structure of the ordered Ta64C phase." Exact orientation means that the plane of the reciprocal lattice lies exactly normal to the incident beam as shown in Fig. 1(b). It should be noted that in exact orientations the angle of reflection is less than the exact Bragg angle 9 so that the reciprocal lattice points lie to the outside of the sphere, Fig. 1(b). This deviation is denoted by the parameter s and in the usual convention4 s is negative for exact orientations and, of course, zero at the exact reflecting position shown in Fig. l(a). In order to avoid secondary reflections from thickness relrods it is advantageous to work in thicker regions of foils. Diffraction from thick regions may also produce Kikuchi patterns and as dis-

"INTRODUCTION Ethyl mercaptan (CH3,CH2SH), a combustible organic liquid of the thioalcohol type, is employed at an odorant in many types of fuel gases and dangerous atmospheres, which in themselves are devoid of characteristic odors, to give warring against leakage of such gases or atmospheres into places where they might constitute health or explosion hazards.Ethyl mercaptan has a penetrating disagreeable odor resembling that of onions, garlic, hydrogen sulfide, or rotting cabbage, that exceedingly low concentrations of it can be detected. The of odorants as a beans of detecting leakage of gases and atmospheres has been investigated extensively by chemists and engineers of the Bureau of Mines, and numerous reports as to its value have been published."

The Bureau of Mines investigated variations in the activation energy for the carbon monoxide reduction of magnetite and ferrous oxide with respect to the state of aggregation of the oxide material. Reduction phenomena can only be satisfactorily explained by including an adsorption step in the reduction process. Summation of the values of the exothermic heat of chemisorption and the variable apparent activation energy for reduction yields a nearly constant value of about 50 kcal for either ferrous oxide or magnetite regardless of whether they are aggregated in pelletized form or coarsely crystalline particles.

This Bureau of Mines study estimates capital investment, operating costs, and selling prices for three underground bituminous mines producing coal with annual production ranging from 1.03 to 3.09 million tons. It is assumed that the mines have a 20-year life. Wages and union welfare payments are considered as of December 6, 1974, under the Bituminous Wage Agreement of 1974, and costs for material and equipment are based on 1975 indexes Initial capital investment ranges from $26,498,500 for the 1.03- million-ton-per-year (MM tpy) mine to $67,173,300 for the 3.09-MM-tpy mine. Total capital investment ranges from $37,437,500 to $92,984,300 for the same mines. Corresponding selling prices for the coal range from $14.83 to $13.26 per ton, assuming a 15-percent discounted cash flow rate of return after Federal income taxes.

From its mining lease at the northern end of Cohens Reef at Walhalla in the North Gippsland Mountains, the Long Tunnel Extended Gold Mining Company produced 13,695 kg of gold, which at 1980 prices represents a value significantly in excess of $200 million. In the period 1885 to 1908 the mine was one of Australia's principal reef gold producers, being top Victorian producer in six of these twenty-four years. Prior and subsequent occupants of the lease produced an additional 160 kg of gold. The Extended and the other mines on Cohens Reef whilst contributing importantly to the financial resource of Victoria, were the sole important support for the substantial town of Walhalla that had grown up around them. When the mines closed, Walhalla gradually reduced from a bustling developed town to the present attractive mountain village so favoured by tuurriSts. The magnesium-rich end of the Mg-Ti phase diagram was investigated. The liquidus, solidus, and solvus boundaries to 1 pct Ti were established. All alloys were prepared by saturating molten magnesium with titanium in a consumable titanium crucible under inert gas maintained at 230 psig. The liquidus of the Mg- Ti system was determined by analysis of dip samples taken from 700° to 1300°C under equilibrium conditions in a pressurized inert atmosphere furnace and by analysis of small ingots rapidly poured and quenched from 1400° to 1500°C. The solubility of titanium in magnesium ranged from 0.018 wt pet Ti at 700°C (0.012 wt pet at 650°C by extrapolation) to 1.035 wt pet Ti at 1500°C. The solidus for compositions ranging from 0.03 to 1.00 wt pet Ti was determined to be 650° ± 1°C by thermal analysis. The titanium solid solubility values ranged from 0.08 wt pet at 350°C to 0.19 wt pet by extrapolation to 650°C. The freezing reaction is peritectic. No intermetallic compounds were found in the system; the phase in equilibrium with molten magnesium saturated with titanium was found to be titanium with magnesium in solid solution. Solid titanium will dissolve at least 1.32 wt pct Mg. PREVIOUS investigations of the Mg-Ti system have shown considerable disagreement on the solubility of titanium in liquid magnesium. Furthermore, the solid solubility of titanium in magnesium has not been well established. Liquidus curves for previous work and for the present investigation are shown in Fig. 1. Aust and Pidgeon1 used a dip-sampling method on molten magnesium held in equilibrium with solid titanium under a protective atmosphere to determine the solubility and found that it ranged from 0.0025 wt pet Ti at 651°C to 0.015 wt pet Ti at 850°C. Eisenreich2 introduced titanium into molten magnesium by means of TiCL4 adsorbed on BaCl2. Ingots were then cast at various temperatures. Making the assumption that only the titanium dissolved in magnesium at the time of casting was soluble in H2SO4, Eisenreich determined the solubility of titanium in molten magnesium to range from 0.003 wt pet at 655°C to 0.115 wt pet at 800°C. Eisenreich also determined the solid solubility of titanium in magnesium to be 0.015 wt pet at room temperature and 0.045 wt pet at 500°C. Since the solid solubility just below the freezing temperature for the bulk of the alloy was much larger than the liquid solubility just above the freezing temperature, Eisenreich concluded that the freezing reaction was peritectic. Obinata et al.3 equilibrated molten magnesium with titanium in hermetically sealed titanium containers which were then furnace-cooled. The titanium content of the magnesium was then determined and found to range from 0.170 wt pet at 700°C to 0.85 wt pet at 1200°C. No intermetallic compound was found in the system. The Armour Research Foundation4 determined two points on the solvus by electrical resistivity methods: 0.00057 wt pet at 200°C and 0.0008 wt pet at 300°C. At higher temperatures, data were meaningless with no trends observable. The authors of this report believed that the lack of significant data at the higher temperatures was due to variations in specimen geometry, although there was no positive evidence to verify this supposition. The present investigation was undertaken to clarify the uncertainty in both the liquidus and solvus of the magnesium-rich end of the Mg-Ti system. EQUIPMENT AND MATERIALS The equipment used in this investigation, with some modifications, was essentially that used by Crosby and Fowler5 in their determination of part of the Mg-Zr phase diagram. The equipment, as modified for this work, is shown in Fig. 2. It consists of a sealed furnace chamber which can be pressurized with inert gas so that melts can be made above the boiling point of magnesium at atmospheric pressure. Melts are made by induction heating in a titanium crucible which, after diffusion of sufficient magnesium into the walls of the crucible to saturate the titanium at the sampling temperature, comprises the solid phase in equilibrium with the molten magnesium. Dip samples may be taken with the sampling tube, or the entire furnace may be tilted so that ingots may be poured into a mold in the side chamber. The principal difference from the earlier apparatus is in the thermocouple, which in the present equipment is enclosed in a protection tube and immersed directly in the melt. The tips of both the thermocouple protection tube and the sampling tube, which dip into the melt, are made of high-purity titanium. Potash, the generic term for a variety of potassium-bearing minerals, ores, and refined products (Table I), owes its importance as an industrial mineral to the potassium requirement of growing plants. The term "potash" originally referred to potassium carbonate produced by the leaching of wood ashes. This process, for which the first U.S. patent was issued in 1790, was the basis for the largest United States chemical industry in the early nineteenth century. The agricultural use of potash soon eclipsed the chemical applications following the German discovery in 1840 that potassium is essential to plant growth. The initial discoveries of large deposits of soluble potassium minerals, essential for agriculture applications, were made in 1857 in Germany and in 1914 in the United States. More than 90% of the 20.4 million short tons K2O produced in the world in 1970 (Eilertsen, 1971 ) was derived from marine evaporite deposits and approximately 90% was used in fertilizers. The potassium content of an ore, mineral, or product is customarily expressed as % K,O, a unit of measure which is neither a natural nor synthetic compound of potassium (1 .O% K is equivalent to 1.2046% K,O) . Geology Potassium, the seventh most abundant element in the earth's crust, is present in silicate minerals in igneous, sedimentary and meta- morphic rocks. Potassium is also a major constituent of many surface and subsurface brines (Table 2). Economic potash deposits are essentially restricted, however, to widespread, thick accumulations of marine chloride evaporite deposits. These deposits yield high grade, large tonnage ore bodies, many of which are amenable to low cost mining and beneficiation. The products from these deposits are, moreover, ideal for use in fertilizer because of the high relative solubility of the potassium chloride and sulfate evaporite minerals. World potash production and reserves for such de- posits so vastly overshadow other sources of potassium that the following discussion will be limited largely to potash deposits in marine evaporites with brief reference to certain potash-rich brines. Approximately 85% of current domestic production is from bedded salt deposits and much of the balance is from brines. Marine evaporite deposits are accumulations of salt minerals deposited in structural sedimentary basins through the evaporation of sea-water or mixtures of seawater and other brines. The salt minerals are precipitated in assemblages and sequences of assemblages as determined by the compositions of the brines and the solubility relations in the salt systems (D'Ans, 1933). The evaporation of seawater normally yields carbonate minerals as the first precipitates (Table 3). Further evaporation and concentration of the brine leads to the precipitation, in succession, of sulfate, chloride, and supersaline mineral assemblages including the salts of potassium and magnesium. The common minerals, ores, and products of potassium deposits are listed in [Table 1]. Many potash deposits contain accessory evaporite minerals together with a variety of detrital and authigenic silicates (Braitsch, 1971).

Mine dewatering is needed in most large open pits but the current practice of using in-pit vertcal wells is often chellenging to implement with good results. In order to address these challenges, SWS collaborated with FMI and innovated a directionally placed mine dewatering well workflow, based on well placement technologies from the Oil and Gas industry. The main aims of the program are to: Maximize hydraulic contact between the ore deposit and production dewatering wells and attain enhanced well production rates from the most important fracture sets. Move the well-heads to permanent areas outside of mine operating areas, removing or reducing the need for in-pit wells and allowing for sustained, prolonged pumping runs times.

The effect of hydrogen on the ductility, c, of SAE 1020 steel at strain rates, i, from 0.05 in. per in. per rnin to 19,000 in. per in. per rnin and at temperature, T, from +150° to —320°F was determined. The ductility surface of the embrittled steel reveals two domains: one in which and the other in which The usual "explanations" of hydrogen embrittlement are in accord with the first of these domains only. THE purpose of this investigation was a fuller A characterization of this of the investigation effects of varying temperature and strain rate on the fracture strain of hydrogen-charged steel. To be sure, it is known that low and high temperatures remove the embrittlement that hydrogen confers upon steels at room temperature,1 * see Fig. la and b, and that high strain rates have a similar effect,'-' see Fig. 2a, b, and c. However, the general effect of these two testing conditions on the fracture ductility of hydrogen-charged steels is not known, i.e., the three-dimensional graphical representation of fracture ductility as a function of temperature and strain rate is not known—only two traverses of the graph are available. The need for such a graph is not pedantic. To demonstrate this point, Fig. 3a, b, and c shows three of many three-dimensional graphs, all possible on the basis of the two traverses at hand. The important point (as will be developed in the Discussion) is that each of them would indicate a different basic mechanism for hydrogen embrittlement. It will be noted that the four types of ductility surfaces in Fig. 3a, b, and c may be characterized as follows: Material and Procedure Tensile tests were made at various temperatures and strain rates on a commercial grade of % in. round SAE 1020 steel in both a virgin state and as charged with hydrogen. The steel was spheroidized at 1250°F for 168 hr to give the unembrittled steel the lowest possible transition temperature. The steel was charged cathodically with hydrogen as follows: The specimen was attached to a 6 in. steel wire, degreased for 5 min in trichlorethylene, rinsed with water, and fixed in a plastic top in the center of a cylindrical platinum mesh anode. The assembly was placed in a 1000 milliliter beaker containing an electrolyte of 900 milliliters of 4 pct sulphuric acid and 10 milliliters of poison (2 grams of yellow phosphorous dissolved in 40 milliliters of carbon disulphide). A current density of 1 amp per sq in. was used which developed a 4 v drop across the two electrodes. All electrolysis was carried on at room temperature. Temperatures for tensile tests were obtained by immersing the specimens in baths of water (+70° to + 150°F), mixtures of liquid nitrogen and isopen-tane (+70° to —24O°F), and boiling nitrogen (-240" to-320°F). Specimens were tested in tension at strain rates of 0.05, 10, 100, 5000, and 19,000 in. per in. per min. The 0.05 and 10 in. per in. per rnin strain rates were obtained on a 10,000 lb Riehle tensile testing machine, the 100 in. per in. per rnin rate on a hydraulic-type draw bench with a special fixture, and the 500 and 19,000 in. per in. per rnin rates on a drop hammer. The fracture ductility of hydrogen-charged steel at room temperature and normal testing strain rates (-0.05 in. per in. per min) is a function of electro-lyzing time, dropping to a value that remains constant after a critical time.'* Under the conditions of • The hydrogen content of the steel continues to increase with charging time even after the ductility has leveled off to its saturated value.' this research the saturated loss in ductility occurred at approximately 30 min, see Fig. 4, and a 60 min charging time was taken as standard for all subsequent tests. After charging the steel with hydrogen, the surface was covered with blisters. These have been described by Seabrook, Grant, and Carney.' The original diameter of the specimen was not reduced by acid attack, even after 91 hr. Results The ductility of both uncharged and charged specimens is given as a function of strain rate in Fig. 5, and as a function of temperature at four different strain rates in Fig. 6. These results are assembled into a three-dimensional graph in Fig. 7. This meeting marks the point at which the long-standing dissatisfaction with the mineral-land laws, the innumerable protests against them, and the many isolated efforts to obtain relief, have developed into a general movement for their revision. That part of the mining industry which is affected by these laws has three general organizations which are able to speak for it: the American Institute of Mining Engineers, the Mining and Metallurgical Society of . America, and the American Mining Congress; all of which are represented in this meeting. In the series of able and thoughtful papers presented here, prominent men have discussed the evils of the present laws and some of the reforms which should be made in them. We all appreciate fully the ultimate value of these studies; but we are also painfully aware of the fact that discussion of this kind has been going on for a quarter of a century without any other effect than a steady increase in the mass of printed records stored on library shelves. In order to get any practical result we Gust induce an indifferent government at Washington to act, and to do this we must unite in pushing some practical plan whereby it may act in a simple and effective way and with the least possible amount of criticism or opposition. The actual question before us is not whether the mining laws are bad; for their evils and absurdities are not only beyond discussion but also beyond the resources of a temperate and dignified vocabulary. It is not what particular changes ought to be made; for it is premature to make . specific recommendations until there is some one to listen to them. Moreover, we all know that there are wisdom and experience enough available, and awaiting that time, for the creation of a code which would be a model to the mining world. The practical question now is, how to obtain the revision; and since the American Mining Congress has of late restricted its attention to this phase of the problem I wish as one of its representatives here to present its views upon the matter.

"Three arc- melted ingots, two of a t itanium-7 percent manganese all oy and the third of unalloyed titanium, were forged and rolled into 3/ 4-inch plate; comparative data were obtained on these processes. The finished plate, part of which was used by the Naval Bureau of Ordnance, was tested for mechanical and other properties.These properties were correlated with sponge purity, forging technique, rolling temperature, and cooling rate from the rolling temperature. For the alloy material, transformation data were obtained, and their age-hardening characteristics were studied. The temperature-impact relationship was established for rolled sections from all three ingots.INTRODUCTIONArc -melted ingots of unalloyed titanium and a titanium-manganese alloy were fabricated into plate under closely controlled conditions by personnel of the Metals and Alloys Section, Physical Metallurgy Branch, Bureau of Mines, College Perk, Md. This program was cosponsored by the Naval Bureau of Ordnance. Bureau of Mines interest lay in correlating the processing procedures with the mechanical properties of the finished plate.Considerable data have been published recently on constitution diagrams and properties of titanium-manganese alloys in genera1. 3/ 4/5/6/ However, little if any correlation between processing schedules and end results has been published, particularly with respect to the present commercial production of the proprietary titanium manganese alloys . Several factors in the production of titanium and titanium alloys have a pronounced influence on the physical and mechanical properties of the finished product: (1) The degree and nature of impurities in the base materials, ( 2) the amount of impurities introduced during the melting cycle and their distribution, (3) the homogeneity of the alloy, (4) forging, (5) rolling, and ( 6) final heat-treating cycles. All of these item£ are important from a metallurgical standpoint in producing the desired end properties in the metal."

"Conventional crushing and grinding is followed by gravity concentration to recover free milling gold. Refractory gold, finely disseminated within the pyrite and arsenopyrite matrix, is recovered by flotation followed by pressure oxidation, cyanidation, and cementation. Non-refractory gold is recovered by cyanidation/CIP processing.A comprehensive test program aimed at improving flotation recovery was initiated in June 1995. Plant testing indicated that flotation tails value could be significantly reduced using mechanical flotation cells. Based on these test results, a flotation circuit that incorporates one of the three existing flotation columns followed by mechanical scavenging and cleaning was developed. Detailed engineering was completed in September 1996. Construction started in February 1997 and was completed in 3 months at a cost of $3.2 million. An on-stream X-ray analyzer was installed to provide real-time arsenic and iron analyses and inferred sulphur value on some of the flotation streams.The new flotation circuit proved to be an instant success. Flotation tails values were reduced by 27%. Final tails solids and solution was reduced from 0.035 to 0 023, and 0.0014 to 0.0006 oz/ton Au, respectively. Based on a mill throughput of 1,620 stpd this decrease in tails value will result in an increased gold production of 7,500 ounces per year. The payback for the project is estimated to be around one year.This paper describes the evolution of the project from bench scale testing to commissioning. Also, the operational and economic impacts on down stream processing are highlighted"

The importance of the so-called "dirty sand" or "conductive solids" problem in electric log interpretation was first stressed in a paper presented in 1949. Since that time the problem has been generally recognized as a most serious one. Several research papers bearing on the theory of the effects of clay contaminants on reservoir rock resistivities have appeared. These papers, while contributing to the theory of the problem, have not offered any practical solution. In the present paper an experimental investigation has been made of the effects of ion-exchange materials on the electrical properties of natural and synthetic porous media. The method used to make synthetic dirty sands is entirely novel and has proved a valuable guide to a better understanding of the properties of dirty sands generally. The effects of ion-exchange materials on both the resistivity and self potential (S.P.) logs have been examined. From the data obtained it has been possible to formulate a simple, practical method whereby electrical logs of dirty sands can be qualitatively interpreted. The method is in all essentials identical to that presently in use for interpreting clean sands. New light has also been thrown on the significance of the term formation factor when it is applied to dirty sands. In 1949 a paper was presented1 which showed that the presence of so-called "conductive solids" introduced considerable complexities into the analysis of the resistivities of dirty sands, i.e., permeable formations containing clay. The reduction of the self potential (S.P.) resulting from the presence of interlaminated shale or interstitial clay in permeable formations has been discussed by Doll.2,3 Noteworthy papers which dilate further upon the effects of shale and clay on the resistivity of permeable formations are those of deWitte,4,5 Berg,6 Wyllie,7 and most recently, Winsauer and McCardell.8 The last authors have endeavored to elucidate the chemistry of the resistivity phenomena in dirty sands in terms of an adsorption hypothesis. Specifically, they attribute the electrolytic conductivity of shales and clays within a permeable formation to an ionic double-layer effect and, for this reason, regard the term "conductive solids" as a misleading one. Some consideration has been given to the effect of dirty sands on the S.P. by Wyllie7 and by McCardell, Winsauer and Williams." Published work to date has cast light only on certain theoretical aspects of the problems created by the presence of clays and shales within permeable formations. ALTHOUGH few changes can be reported in educational methods at the mineral technology schools during 1940, other events have taken place of direct interest to, and that will have a profound effect upon, mineral industry education on the United States. In the erection of new buildings, some notable changes have been made on the campus, The U.S. Bureau of Mines Experiment Station at the University of Utah, Salt Lake City, was dedicated on May 20. The construction and equipment of this building cost between $300,000 and $400,000. It will be the general headquarters for the metallurgical division of the Bureau and will have a pronounced effect upon the mineral industries research program at the University.

his report is going to point to that the effect of the mining dewatering on the area of Transdanubian Mountain Range has reached a phase when the cost of dewatering and providing damages can seriously jeopardize a profitable mining operation. Up to 60s the interest of mining had been dominant only, but since the begining of 80s the conceptions of industrial development and the value of enviroment protection have been changed. The natural karstwater balance of the TMR can be presented (Figure Land 2.) and the present situation of it points to that the descharge has exceeded the reacharge in 50 %. During the past 10 years the decreasing of the stored karstic water resourches was 1.1 km . The decreasing of the spring yields reached 80-82 % of the natural ones. (Figure 3-4.). Conservation of the present discharge may lead up to the totaly drain of the karstwater regime. A computer aid control system had been to develop by the bauxite mining and using of it the yield of dewatering decreased and a moderate rising of the water table has started since 1986. A recunstruction was made at the biggest thermal-medicinal lake its springs have had their source in 40 meter depth inside a spring cave at the bottom of the lake. Earlier the spring yield decreased due to the dewatering activity of mines. As a result of using of the CAC system and the providing works at the thermal sprino,the karstwater balance has got in the state of equilibrum, the first cost was decreased and spring yield was increased and the enviromental limits were adverted from the way of mining.

"It has been recently suggested that the exceptionally high antitumor and antibacterial activity of natural fredericamycin A (FMA) is related to its ability to spontaneously generate the superoxide anion (02-) and hydroxyl (""OH) radicals in aerobic solutions [Hilton, B. D., Misra, R., & Zweier, J. L. (1986) Biochemistry 25, 5533]. With a view to understand the mechanistic details, attempts were made to reproduce earlier electron spin resonance (ESR) evidence for the oxygenated free radical formation in well-aerated solutions of natural FMA in dimethyl sulfoxide and dilute H 20 2• Little or no evidence was obtained for the formation of the 0 2- and methoxy (""OCH3) radicals, while the detected formation of the 'OH and methyl ('CH3) radicals was attributable largely to mechanisms not involving FMA. These results thus reopen the T question regarding the mechanism of its exceptionally high tumoricidal-bacteriocidal activity. This paper reports on an electron spin resonance (ESR) investigation of the formation of oxygenated free radicals in solutions of natural fredericamycin A (abbreviated as FMA) which has recently been reported to be a highly potent anti¬tumor antibiotic (Pandey et al., 1981; Warnick-Pickle et al., 1981). The impetus for this undertaking was provided by recent reports (Hilton et al., 1986; Stinson, 1986) suggesting that the strong tumoricidal activity with high therapeutic—toxic index of FMA was related to its ability for the spontaneous generation of superoxide anion (02-) and hydroxyl (OH) radicals. For example, the suggested mechanism for the formation of 02- by FMA was (Hilton et al., 1986)FMA-OH + O2 FMA-O + H+ + 02-Here FMA—OH is simply the FMA molecule emphasizing the —OH moiety which becomes FMA-O, the free radical rep¬resenting the oxidized form of the drug. While some other highly effective anticancer drugs in current clinical application, such as bleomycin, are also thought to exert their antitumor effects via the formation of reactive drug-based or oxygenated free radicals (Carmichael et al., 1985; Gianni et al., 1983), FMA was considered unique in that only in this case the free radical formation was thought to be spontaneous, rather than through a metal- or an enzyme-activated process (Hilton et al., 1986; Stinson, 1986). Thus it was suggested (Hilton et al., 1986; Stinson, 1986) that FMA could be the forerunner of a new class of anticancer drugs. However, we noted that while the hypothesis seemed plausible, the reported ESR spectral evidence appeared to be ambiguous. Because of the high significance of the earlier conclusions (Stinson, 1986), we undertook a systematic ESR study of the radical formation in FMA solutions, and as discussed below, our conclusions disagree with those reported earlier."
