"SUMMARYElectron spin resonance (ESR) measurements provide direct evidence for the involvement of the glutathionyl radical, as well as an isolable Cr(V)-glutathione intermediate in the Cr(VI) glutathione redox reactions. These results indicate a new mechanism for the reduction of Cr(VI) by glutathione in relationship to cellular environment and suggest that synergistic reactions of Cr(V) and GS· might be responsible for the (unexpected) increase in the Cr(VI)-induced DNA-strand breaks at high glutathione levels.INTRODUCTIONWhile the reduction of Cr(VI) by glutathione (GSH) is thought to be a critical' step in the pathogenesis of the Cr(VI)-exposure related carcinogenesis1- 4, the exact reduction mechanism is not yet understood. The following reaction steps have been suggested :(A) Initial step:Cr04 2- + GSH - GSCrOa - (1)(B) At high GSH levels:GSCroa- + GSH - Cr( IV) + GSSG (2)(C) At low GSH levels: GSC rO a - - Cr ( V ) + GS ·(3)Thus far, however, in these reactions, the GS· radical has eluded spectroscopic detection, while the evidence for the Cr(V) formation has been reported positives as well as negative2·. Her~ we present spin-trap ESR evidence for the formation of the GS· radical, and a definitive ESR detection of an isolable cr(V)-GSH intermediate in Cr(VI)-GSH reactions.MATERIALS. AND METHODSESR spectra were obtained at X-band (-9.7 GHz) using a Bruker ER200D ESR spectrometer. The magnetic field was calibrated with a self-tracking NMR Gaussmeter (Bruker, Model ER035M) and the microwave frequency was measured with a Hewlett-Packard (Model 5340A) frequency counter. The spin probes, a-(4-pyridyl-1-oxide) -N -tert-butylnitrone (4-POBN) and 5,5-dimethyl~1-pyrroline-1-ox1de (DMPO) were purchased from Aldrich, K2Cr201 from Fisher and used without further purification. Seals are barriers constructed in underground coal mines throughout the United States to isolate abandoned mining panels or groups of panels from the active workings. Historically, mining regulations required seals to withstand a 140-kPa (20-psig) explosion pressure. However, the Mine Improvement and New Emergency Response Act (“MINER Act”) requires the Mine Safety and Health Administration (MSHA) to increase this design standard by the end of 2007. This report provides a sound scientific and engineering justification to recommend a three-tiered explosion pressure design criterion for new seals in coal mines in response to the MINER Act. Much of the information contained in this report also applies to existing seals. Engineers from the National Institute for Occupational Safety and Health (NIOSH) examined seal design criteria and practices used in the United States, Europe, and Australia and then classified seals into their various applications. Next, the engineers considered various kinds of explosive atmospheres that can accumulate within sealed areas and used thermodynamic calculations and simple gas explosion models to estimate worst-case explosion pressures that could impact seals. Three design pressure-time curves were developed for the dynamic structural analysis of new seals under the conditions in which those seals may be used: unmonitored seals where there is a possibility of methane-air detonation or high-pressure nonreactive shock waves and their reflections behind the seal; unmonitored seals with little likelihood of detonation or high-pressure nonreactive shock waves and their reflections; and monitored seals where the amount of potentially explosive methane-air is strictly limited and controlled. Figure 1 is a simple flowchart that illustrates the key decisions in choosing between the monitored or unmonitored seal design approaches and the three design pressure-time curves. For the first condition, an unmonitored seal with an explosion run-up length of more than 50 m (165 ft), the possibility of detonation or high-pressure nonreactive shock waves and their reflections exists. The recommended design pressure-time curve rises to 4.4 MPa (640 psig) and then falls to the 800-kPa (120-psig) constant volume (CV) explosion overpressure. For unmonitored seals with an explosion run-up length of less than 50 m (165 ft), the possibility of detonation or high-pressure nonreactive shock waves and their reflections is less likely. A less severe design pressure-time curve that simply rises to the 800-kPa (120-psig) CV explosion overpressure may be employed. For monitored seals, engineers can use a 345-kPa (50-psig) design pressure-time curve if monitoring can ensure that (1) the maximum length of explosive mix behind a seal does not exceed 5 m (16 ft) and (2) the volume of explosive mix does not exceed 40% of the total sealed volume. Use of this 345-kPa (50-psig) design pressure-time curve requires monitoring and active management of the sealed area atmosphere. These design pressure-time curves apply to new seal design and construction.

"Summary: In this report the differential regulation of NF-KB and nitric oxide (NO) was investigated in the mouse macrophage cell line RAW 264.7 following exposure to a mineral dust (silica) and /or an endotoxin (Lipopolysaccharide, LPS). The results indicated that silica and LPS can significantly induce the activation of NF-KB as well as elicit enhanced production of NO in RAW 264.7 cells as part of an early inflammatory response mechanism. A 24-hour time-course study showed that NO release from these cells continued to increase following the initial stimulus by LPS or silica. In contrast, activation of NF-KB was maximal at 6 hours and then showed a steady decline to 24 hours. The production of NO was suppressed by protease inhibitor and antioxidant, both of which block the activation of NF-KB. Surprisingly, the use of an NO synthase inhibitor resulted in an enhancement of NF-JCB activation. These findings suggest that NO produced in macrophage cells in response to an inflammatory stimulus like silica or LPS my be linked to a negative feedback role on the activation of NF-KB. It is now accepted that nitric oxide (NO) plays a pivotal role in the inflammation response process when macrophage cells are activated by a pro-inflammatory stimulus (l). Production of NO is dependent on the activity of the enzyme nitric oxide synthase (NOS). Up until now, at least three distinct genes have been described which encode different isoforms of NOS. NOS has been cloned from a variety of different mammalian cells, including constitutive neuron NOS, constitutive endothelium NOS and inducible NOS (2). Macrophages appear to be the principal cellular source for the inducible form of NOS (iNOS) activated in response to an inflammatory producing event. The mechanisms subserving the regulation of the iNOS gene and NO production have been under intense investigation in recent years (3-5). Several binding sites for transcriptional factors have been identified in the promoter region of iNOS gene, including NF-lCB, AP-1, NF-IL6, Oct-I to cite a few (5). NF-KB appears to play a"

In 1984, at the request of the Bureau of Land Management, the Bureau of Mines studied 29,020 acres of the 43,165-acre Terry Badlands Wilderness Study Area (MT-024-684) in order to evaluate its mineral resources. Within this report, these 29,020 acres are referred to as the study area. Located in Custer and Prairie Counties, MT, the study area is about 2 miles northwest of Terry. The study area is underlain by essentially flat-lying units of the Fort Union Formation of Paleocene age with some Quaternary alluvial deposits in stream valleys. No mines or prospects were found in the study area. Bentonitic clays and lignite coal units were examined and sampled; however, no clay resources or coal reserves were identified. Estimates of coal resources will be reported by the U.S. Geological Survey in a joint U.S. Geological Survey-U.S. Bureau of Mines publication. Gravel along the Yellowstone River, a source of agate for private collectors, could also be used as sand and gravel aggregate although material on the south bank of the river is closer to population centers and road systems. Clinker found in the study area could be a source of road metal, but is not well situated in relationship to areas of use. No geothermal resources have been identified although water suitable for direct heat applications may underlie the study area. All the area is leased for oil and gas exploration.

Develop a technology for producing high-quality hake graphite from the steelmaking waste known as kish. Approach Treat the waste kish as an ore and apply mineral processing techniques of physical beneficiation and chemical purification to produce a range of graphite products com¬petitive with the natural mineral. Background Graphite is a naturally occurring form of carbon with a unique combination of properties that make it indispensable in a very wide range of industrial and consumer applications. It has high electrical and thermal conductivity and remains stable to extremely high temperatures It has a low coefficient of friction and is nearly impervious to corrosion. The steel industry is dependent on graphite in clay-, magnesia-, and alumina-bonded refractories for crucibles, bricks, facings, and coatings. Electrical uses include graphite brushes in motors and generators, and electrodes in batteries. The transportation industry needs graphite for clutch and brake linings, and all industries depend on graphite in seals, gaskets, lubricants, and coatings. The consumer sees graphite in composite plastics and in the common pencil. As with all natural minerals, the availability of graphite is diminishing and costs are rising as quality decreases.

Develop a reliable, portable device for non-destructively testing the support integrity of a friction rock support (commonly referred to as a Split Set ). Approach The friction rock support, used to support mine roof and ribs, is a slotted tube driven into a drilled hole of smaller diameter. Upon installation, the tube is compressed radially. The amount of compression is a function of the hole diameter, the strength of the host rock, and the properties of the split set itself. A simple lightweight self-contained device was developed to determine the pull-out resistance of the friction rock supports in-situ, based on measurements of the tube's deformation. The tester measures the com-pressed internal volume of the slotted tube, which has been found to provide an estimate of the support's resistance to pull out. How It Works The tester consists of a pump and expander sections. The expander sections are made from an elastomer membrane which is stretched over two cores, each one inch in diameter and twelve inches long. The expanders are connected by a flexible coupler so that they can fit any alignment of the support.

The Bureau of Mines developed by core drilling the Cove Meadow copper property, also known as the Patterson property, from May 6, 1949, to August 6, 1949. It is in the Cove Meadow mining district, northwestern Humboldt County, Nev., 102 miles northwest of Winnemucca. The purpose of the work was to determine possible reserves in a mineralized shear zone in andesite that has been partly explored by several surface cuts and two shallow underground workings. Production from this zone to August 1, 1,949, was 98 tons of hand-sorted copper ore assaying 5.1 percent. The development work performed by the Bureau of Mines comprised a study of the geology, mapping the area, sampling the outcrops, and exploring by diamond drilling the shear zone adjacent to the fault. Seven holes were drilled for a total of 1,246 feet. Mineral reserves indicated by the project are confined to a length of 500 feet along the strike.

Several sets of questions and answers on the fundamentals of first aid to the injured, based on the Manual of First-Aid Instruction of the United States Bureau of Mines, are now in use in various parts of the United States. Persons experienced in first-aid instruction believe that a standardized set of questions and answers will assist materially in preparing men for examin- ations conducted by the Bureau for first-aid certificates. Inquiries as to whether a set of standardized questions and answers is available have been made frequently by those training men for the Bureau of Mines examination for certificates. It is believed that the following list of 171 questions and answers com- piled directly from the manual will be useful to company officials and em- ployees who are engaged in cooperative first-aid training to qualify for Bureau of Mines certificates. The list may also be used by field men of the Bureau in training instructors preparatory to a cooperative training campaign and in examining classes or individuals for first-aid certificates. The page references in parentheses refer to pages in the Manual of First-Aid Instruction.

"One of the most serious long term health problems associated with underground,mining is Coal Workers Pneumoconiosis (CWI'), more commonly known as Black Lung Disease. It occurs in some coal workers, normally after 15 or more years of exposure to coal mine dust, and leads to a variety of respiratory problems. Two types of pneumoconiosis are recognized simple CWP, which has been estimated to affect 6.9 percent of miners and is a condition that does not impair the miner's ability to work nor reduce life expectancy; and a more severe form of black lung (called progressive massive fibrosis) that occurs in about one percent of the cases and results in severe lung damage.Although the relationship between coal dust and CWP has long been recognized, detailed knowledge of the relationship is insufficient for setting up authoritative guidelines to reduce the health risks for those employed in dusty,underground environments. CWP is not only a health problem for workers that causes considerable distress, it is also a very expensive problem the total annual costs associated with CWP approach two billion dollars.As part of its efforts to combat CWP, the U.S. Bureau of Mines in 1983 established a Generic Mineral Technology Center to carry out basic research into all aspects of respirable coal dust:— control of dust generation — the behavior of dust in the mine — characterization of dust particles interaction of dust and lung— the relationships among dust generation and mobility and the mine environment, geology, and seam characteristics,Penn State, West Virginia University, the University of Minnesota, and the Massachusetts Institute of Technology are involved in this research, which is administered from the Department of Mineral Engineering at Penn State.Our work is one of the seven projects to be conducted at Penn Slate. We are concerned particularly with the characterization of dust and its relationships with the mine environment, and our main goal is to establish standard procedures for describing and measuring these variables.Coal DustCoal dust is a very complex material that varies greatly in chemical and mineralogical properties. Moreover, it is not the only constituent of respirable dust in the mine. When a coal seam is mined, it is inevitable that rock particles other than coal are included in the material. This rock comes from the mine roof, the floor, and the partings and inclusions within the coal scams. Since the roof and floor strata, the partings and inclusions are often stronger than the coal, mining them frequently causes more dust than mining the coal itself. One of the objectives of our research is to find the relationships between a specific coal's characteristics before it is mined and the respirable dust characteristics after it is mined. We are particularly interested in the size distribution of particles, the chemical properties, and the mineralogical properties."

In August 1943 the Bureau of Mines completed seven churn-drill holes in an area of old mine workings near Linden, Wis., formerly known as the Argall mine. The work was undertaken as part of a general investigation by the Bureau of the zinc and lead producing area of southwestern Wisconsin at a time when special effort was being made to increase the production of metals necessary for war. The area was selected for investigation on the basis of its favorable geologic structure, past history, and apparent mineralization.

The response of minerals in magnetic separators is in accordance to dynamic rather than static magnetic susceptibility values . The dynamic value varies with respect to both passage speed through the separating field and time of exposure in a preliminary treatment field . Tests on 16 minerals and solids have demonstrated that both of these factors affect different minerals in different ways .

A sedimentation method involving the use of the Palo-Travis apparatus was found satisfactory for rapid determination of the size distribution of coal particles ranging from 10 microns in diameter to those passing an 840-micron opening (20-mesh sieve), The method is most useful for comparing the size distributions of samples of pulverized coal where the actual size of the particles is not required and is applicable to sieve and subsieve sizes of a wide variety of coals and chars that can be wetted and are heavier than water. An analysis, including all calculations, can be completed in 1/2 to 10 hours, depending on the time required for the finely divided coal to settle. A typical analysis requires about an hour, making this a rapid, precise method for routine size-distribution analysis of powdered coals. When used with coal particles smaller than 400 microns, the method is precise within 15 microns for wide variations in test conditions and within 5 microns for identical test conditions. The precision decreases as particle size increases, becoming unsatisfactory for particles larger than 400 microns (40-mesh). For precision within 15 microns, the samples must be equal in weight. Greater precision is obtained with 4-gram samples, but samples as small as l gram may be used.

The U.S. Bureau of Mines and the United States Steel Corporation evaluated the progress of methane drainage at the Oak Grove, AL, degasification pattern. Coalbed pressures were monitored between December 1981 and November 1985. Formation pressures, in the lower bench of the Mary Lee Coalbed, were reduced from 400 lbf/in2(ga) in 1977 to 50 lbf/in2(ga) in November 1985. Gas contents of coal cores obtained from monitor wells were compared with coalbed gas contents measured in 1976. Data indicated a 48- to 56-pct reduction within the pattern and a 29-pct reduction at a point 500 ft outside the pattern. The effect of the advance of a section of the Oak Grove Mine on changes in gas and water production rates in the pattern was evaluated. Gas production increases were noted in 12 wells; water production decreases were noted in 6. The times of initial influence indicate the presence of a zone of high permeability within the pattern. This zone correlates well in orientation and location with a fracture zone mined through at the mine. The existence of such fracture zones could have a significant effect on productivity of individual coalbed methane wells.

As part of the work of serving as Chairman of Subcommittee 3, Health and Industrial Requirements, of the American Society of Mechanical Engineers' Committee on Air-Pollution Controls, the author assembled the following information on accepted maximum permissible concentrations of air pollutants from the standpoints of health, damage to vegetation, damage to property, and requirements of industrial processes. For the past few years the Applied Physics Branch has been investigating, experimentally, the generation and propagation of strain waves in rock. The strain waves are produced by the detonation of a high-velocity explosive placed in a small cavity in the rock. Strain gages mounted in the solid rock surrounding the cavity record the strain pulses propagated away from the source. Numerous strain-wave pulses have been recorded in different rock types for charges of various sizes and at various distances from the charge. Analysis of these records has shown, among other things, that there is associated with the strain wave a characteristic pulse width. This pulse width is dependent on the rock type and the distance from the shot point to the gage, but is apparently independent of the charge size or cavity size. Recent published literature on the generation and propagation of seismic waves indicates that the characteristic frequency of an elastic wave generated by an explosion should be a function of the rock type and the size of the cavity created by the explosion. Also, internal friction in the rock would produce absorption of high-frequency components, which would cause continual change in the shape of the pulse propagated. As the experimental data did not verify the expected change in pulse shape with cavity size, a theoretical investigation of the vibrations associated with a spherical cavity in an elastic medium was undertaken.

It is now widely accepted that horizontal stress is present nearly everywhere underground. Often it is more severe than the vertical stress. It is a major cause of roof control problems in underground mines. The AHSEM computer program contains two modules to improve mine layouts for control of horizontal stress. For development, the user specifies the orientations of the maximum horizontal stress, the entries, and the crosscuts. Angled crosscuts may be used, and the angles may be different for the right- and left-hand crosscuts. AHSEM then shows the user how the stress condition would vary as the entry and crosscut orientations are changed. The second module is for longwall mining. Severe stress concentrations can make headgate conditions hazardous when the entries are improperly oriented. AHSEM shows whether a particular longwall orientation and extraction sequence places the headgate corner in a stress concentration or in stress relief. It also estimates the magnitude of the stress concentration.

The theory of electromagnetic fields indicates such fields could be used to detect and locate trapped miners. To be useful, the hardware of the system must meet a number of requirements, including small size, intrinsic safety, and rugged construction. Such hardware has been built, and the system has been tested by the Bureau of Mines and its contractors. These tests indicate that the electromagnetic method provides a practical means to locate miners in emergencies.

"INTRODUCTION The Miscellaneous Minerals Division of the War Production Board requested the Bureau of Mines to investigate the Gallatin corundum property as a possible source of corundum, which was in short supply due to the war. The Gallatin corundum property, also known as the ""Fuller mine"", the ""Montana Corundum mine"", and as the ""Old Qorundum mine"" was first examined by a Bureau of Mines engineer 2/ in July 1943. Bureau of Mines work on the property started October 11, 1943, was recessed May 5, 1944, and was resumed in October 1944 when the diamond-drilling program started. The project was completed February 7, 1945. The data obtained are given in this report.The Federal Geological Survey made a reconnaissance survey in the spring of 1943 and mapped the area the following autumn. The results of these surveys are contained in a memorandum report prepared in December 1943.ACKNOWLEDGMENTSIn its program of investigation of mineral deposits, the Bureau of Mines has as its primary objective the more effective utilization of our mineral resources to the end that they make the greatest possible contribution to national security and economy. It is the policy of the Bureau to publish the facts developed by each project as soon as practicable after its conclusion. The Mining Branch, Lowell B. Moon, chief, conducts preliminary examinations, performs the actual investigative work, and prepares the final report. The Metallurgical Branch, R. G.
