"With a view of elucidating the role of glutathione (GSH) in the biochemical pathways of the chromate-exposure related carcinogenesis, we carried out electron spin resonance (ESR) spectroscopic investigations of the chromate-GSH redox reactions. The ESR measurements, employing spin-traps, provide evidence for the involvement of the glutathione (GS•) radical, as well as an isolable Cr(V)-glutathione intermediate. These results indicater a new mechanism for the reduction of chromate by GSH in cellular environment and help understand the (unexpected increase in Cr(VI)-induced DNA strand breaks at elevated GSH levels.We report here indirect electron spin resonance (EGR evidence for the involvement of the glutathionyl (GS) radical and direct evidence for a Cr(V) intermediate in the reactions of glutathione ¬(GSH) with chromate for the first time.. This work was undertaken to clarify the biochemical pathway of the reduction of chromate by GSH, since this reduction process is thought to be a critical step in the pathogenesis of the chromate-exposure related carcinogenesis. This follows from the literature reports which show that (a) Cr(VI)compounds have been proven to be carcinogens in laboratory animals as well as in human studies2-4, (b) Cr(VI) and not Cr(III) compounds are mutagenic 5-7, (c) Cr(VI) and not Cr(III) compounds can penetrate cell membranes 8.9 (d) the final state of Cr(VI) in cellular reductions is Cr(III)', (e) by itself chromate (or Cr(VI)) cannot be directly mutagenic since Cr(VI) is known not to interact with isolated DNA under physiological conditions 10, suggesting the significance of Cr(VI).reduction processes. One of the major reductants in cellular environments is thought to be the GSH, both outside and inside the ce11s 11-13. Other evidence for the role of GSH in the chromate toxicity is seen from recent studies showing that exposure of hamster cells to non-toxic levels of added selenite increases the levels' of GSH as well as the Cr(VI)-induced DNA strand beaks 14, and thatsuch DNA strand breaks in hepatocytes also change in direct proportion to their GSH content 15,16. These observations wereinterpreted as implying that the reduction of chromate by GSH, to some reactive intermediate is an important step in the chromatecarcinogenicity 15,16. "Inhaled dust particles of appropriate size contact cells of the alveolar region.' The acute response to coal dust appears to involve a variety of cell types, including alveolar epithelial cells, interstitial fibroblasts, both resident and recruited macrophages, and cells of the immune system. Direct and indirect effects of dust, which may be mediated by soluble factors,'' can initiate acute and long-term lung injury. While it appears that epithelial cell growth and differentiation, along with integrity of the epithelial basement membrane, may play a role in the long-term response to dust exposure, little is known of the role the alveolar epithelium may play in the progression of injury. The broad hypothesis that underlies these experiments is that short- and long-term consequences of coal dust exposure involve changes in structure and function of the alveolar epithelium and the underlying extracellular matrix. Recent studies show that coal dusts exert direct effects to modify pulmonary epithelial cell-extracellular matrix interactions in vitro.3EXPERIMENTAL PROCEDURESType II cells isolated on day 0 by density centrifugation and differential attachment4 were plated overnight in D MEM containing 10% fetal calf serum before exposure to anthracite dust PSOC 867 sonicated into culture medium (750 ug/mL).3 Synthesis of extracellular matrix proteins was determined on day 3 of primary culture, as detailed elsewhere. 5 Cells were removed from extracellular matrix by extraction with NH4OH and high salt. Extracellular matrix synthesis and fibronectin content were evaluated based on radiolabeling with 3 H-leucine and on Western blot analysis, respectively. Additional analytical methods and calculations are detailed elsewhere.3-5"

Federal law mandates that all miners receive safety and health training when first entering the mining industry and at least 8 hours annually thereafter. Although the mining industry has historically relied on an experiential master-apprentice model for training new employees, the formal safety training provided to miners is generally done in a classroom, with mixed results. In a series of stakeholder meetings held by NIOSH across the West in 1997-2000, trainers identified significant gaps in materials that were available to teach new and experienced miners. They asked whether NIOSH could develop effective materials to fill these perceived gaps, as well as make training more effective for those who were required to attend, but who often refused to be attentive. Adult learning theories, social learning theories, mentoring, and storytelling were all employed in the development of a series of safety training videos that have become very popular in the mining industry. This Report of Investigations describes the theoretical frameworks that were used, the process that was developed to produce the videos, and the results of the evaluations as to whether or not they were effective in training new and experienced miners.

"The diesel particulate matter (DPM), a constituent of the diesel engine exhausts, is known to be a potential carcinogen. 1- 3 Consequently, suitable methods to control/eliminate DPM have ·been under investigation in recent years. The DPM can be controlled by using fibrous ceramic traps. However, clogging of traps by DPM requires the impractical periodic trap replacement. Following the recent work by Garner and Dent,4,5 we are investigating the in situ use of microwaves to regenerate the ceramic traps, thereby eliminating the trap replacement step in this process. The use of microwaves instead of conventional heating for trap regeneration has the advantage of selective heating of the DPM instead of the trap, if the trap absorbs insignificant microwave power relative to the DPM. Also, the oxidation characteristics of the DPM are needed to understand fully this regeneration process.The rate of energy absorption P per unit volume of a sample is given byP = wE"" |Eo|2 • (1 )where w is the angular frequency, |E0|2 is the intensity of the microwave radiation, and E” is the imaginary part of the dielectric constant (E = E’ + iE”) . Thus measurements of E11 for DPM and ceramic trap material are needed to determine their capacities for microwave absorption. In this paper, we report these measurements carried out at · a frequency of 8.7 GHz. Following microwave absorption, the DPM is expected to oxidize. The oxidation characteristics of the DPM have been investigated using Fourier transform infrared (FTIR) spectroscopy of the evolved gases CO2 and CO and using thermogravimetric analysis (TGA). Finally, some information on the structure of carbon in DPM has emerged from the analysis of its x-ray diffraction spectra. Results of these experiments are reported in the following pages."

"At the beginning of 1920 a general feeling of optimism was noted in many talc-producing districts. The industry seems to have recovered from the temporary depression of the first half of 1919 and many mills are running at capacity production with their entire output for 1920 contracted for. The imports of talc for 1919 were only slightly larger than in 1918 by weight and probably no more by value. Thus foreign competition has not begun to be seriously felt. Several new companies in this country have been organized, some to operate new deposits and others to continue the work done by others.An interesting phase of the present situation is the possibility of increased use of talc in paint. There is a shortage of barium sulfate used as,paint filler. Since, for some uses, talc is considered second only to BaSo4 as a filler for paint, it is possible that part of the shortage could be relieved by an increased use of talc.A series of physical tests upon all pigment materials used in the manufacture of paints is being conducted for the Paint Manufacturers Association of the United States. It is important that all types of talc be represented in these tests. Producers of finely ground talc, 200 mesh and finer, are urged to submit samples of at least two pounds each of these grades that are available to paint makers with trade name and quality data, to the Institute of Industrial Research, 19th & B Streets, Washington, D. C., where the tests are being conducted.The Talc Products Company, Inc., Washington Building, Los Angeles, California, has recently been organized to develop a large body of talc one mile north of Riggs, San Bernardino County, California, This deposit contains a compact, white talc which can be sawed into crayons. The company's mill, at Los Angeles) is equipped with a unit of the Raymond system of grinding and air separation, and pencil sawing machines. It is reported that the pencils are being successfully used in Pacific Coast ship yards.The mining of talc where no mill is owned by the operator and the consequent sale of talc in its crude massive state has not proved profitable in a number of cases recently noted. Under exceptional circumstances, where mining costs are low and the product can be sold to a mill near the mine, the operation maybe profitable, but this condition is not common. One mine of this type has recently closed down, at least temporarily; but a second, more fortunately situated, in another district, is continuing operations, although plans are being made to erect a mill in the early spring. Most satisfactory results are ordi¬narily obtained where the mine and mill are owned by the same company.The sale of the Balmat mine of the International Pulp Company, near Gouverneur, N. Y., to J. H. McLear, Gouverneur, and H. G. McLear of New York, has been reported. It is stated that a large part of the property has been already leased to the Sylvia Lake Company, who will operate it. ""Cement, Hill, and Quarry""* states that ""the first mining done on this property was for silver and lead, but it developed into a good zinc producing property and later it was worked for its talc deposits. It has passed through several hands, the American Pulp Co., the Union Talc Co., and the International Pulp Co., all having had it in successive periods since 1891."""

A spectrophotometric method for the determination of small amounts of copper in tungsten metal powder using the principle of solvent extraction has been developed by the Federal Bureau of Mines. The metallic sample is dissolved in hydrogen peroxide, tartaric acid added, the pH adjusted, and the excess peroxide destroyed by boiling. Copper is reduced to the cuprous state, and the reagent neocuproine (2,9-dimethy1-1,10-phenanthroline) is added to form a colored complex with the copper. This colored complex is then extracted into chloroform and the absorbance of the separated chloroform layer measured on a spectrophotometer. Copper contents as low as 1 p.p.m. (parts per million) can be determined. The method is limited to finely divided tungsten powder. The interest of the Bureau of Mines in developing more sensitive methods for the determination of trace impurities in high-purity tungsten prompted an investigation of the possibility of using solvent extraction-colorimetric methods. The organic reagents now commercially available make possible many extremely sensitive colorimetric methods for the determination of metallic ions. Extraction of the colored complex into a small volume of organic phase can be used to further increase sensitivity (4) .4/ This general approach has the added advantage that no special equipment beyond that normally found in an analytical laboratory is required.

Ventilation-air-heating plants are in use at several iron mines on the Menominee Iron Range, Iron River, Mich. Many of the mines are exceptionally wet; during winter months, when fresh air is forced down the wet air shafts into the mine, it is usually necessary to heat the fresh air to above freezing so that the openings will not be closed by ice. The problem of ice closing air shafts has been solved by the practice of reversing the air current before the openings have become completely closed. This procedure involves drawing warm air from the mine up the usual down-draft air shaft. The M. A. Hanna Co. has found, at some of their mines on the Menominee Range, that it is more convenient to heat the below-freezing air rather than reverse the air current. Intermittent freezing and thawing of ice that has formed in the air shaft tends to loosen the wall rock, requiring major shaft repairs. The openings are kept free of ice the year around by heating the fresh air during periods of below-freezing weather. Table 1 shows the monthly minimum moan, maximum mean, and mean temperatures for the Iron River area. Under normal conditions of temperature, the furnaces are fired in November and usually are not allowed to go out until April of the following year.

"The United States Bureau of Mines is cooperating with the State of Utah and the City of Salt Lake in an investigation of methods of abating the smoke nuisance in Salt Lake City. In the course of this work samples of the atmosphere have been collected and analyzed in an attempt to determine the part played by sulphur dioxide or smelter gases in polluting the air at Salt Lake City.A fairly wide-spread opinion exists in Salt Lake City that the smelters contribute appreciably to the smoke nuisance. The gases discharged from the smelter stacks are believed (1) to float over the city in a heavy blanket which prevents the smoke from rising, and (2) to unite with the water vapor in the air to form corrosive fogs injurious to health and property.The first of these theories seems untenable. If the gas did not mix with air it would sink, owing to its heavier specific gravity, and form a blanket an the earth. Actually, the gas diffuses rapidly in air and any part reaching Salt Take must he thoroughly diluted. Perhaps sulphur dioxide in sufficient concentration might aid in fog formation. The more obvious nuisance, however, would be the injury to health and vegetation.The Garfield smelter is 16 miles west of the city, The Murray smelter 8 miles south, and the Midvale smelter 12 miles south. Under normal operating conditions, a relatively small amount of solid matter is discharged from these stacks. The toxic gaseous constituent of the discharge is sulphur dioxide, it is being discharged from the smelters the year round. If they are a factor in the city's smoke problem, the concentration of this gas in the air of the city should be appreciable and should increase toward the smelters, the con centration being highest with a south or a west wind."

Until man was able to produce light, his working day was pretty much limited to daylight hours. Underground miners have a special problem because in a mine it is just as dark during daytime as it is at night. The first devices used for mine lighting were torches and open flame 011 lamps. Torches made from twisted grass and dipped in tallow or pine pitch would burn for a while, but they made a lot of smoke and probably went out when they were needed most. During the 15001s, miners lamps consisted of a small metal dish with a hook attached to hang it up. The saucer-shaped dish was filled with animal or fish oil and a wick made of twisted fibers was laid In the 011 and hung over one edge where it would burn. This lamp would burn longer than a torch, but the amount of light cast by the pale, flickering flame was probably only a little better than no light at all.

Fly ash from four sources and in certain proportions was shown to assist chemical coagulation of turbid water and settling of chemically induced floc. Fly ash reduced the time required to form the first visible floc, promoted more uniform flocs about 2 mm in diameter, and increased the mean settling rate of floc particles. Also, the addition of fly ash produced denser sludges than those obtained with alum alone. Final turbidities were independent of fly ash particle size and carbon content. Optimum proportions of fly ash additive are given.

Samples taken from the few mineralized exposures justified further investigation of the property. Over 5 million lb of chromium, 1 million lb of nickel, and 90,000 lb of molybdenum are lost annually in the dusts generated in the production of stain-less steel. Additional losses of these metals in the form of mill scale and grinding swarf may be double the dust losses. This Bureau of Mines report describes a process for recovering these and other metals in the form of an alloy steel suitable for recycling. Zinc and lead, when present in the wastes, are recovered as byproducts in the form of oxide fumes. The laboratory-scale process (10-to 25-lb charges) recovers up to 95 pct of the chromium, 100 pct of the nickel, and 100 pct of the iron. The resultant alloy contains from 10 to 20 pct chromium, 6 to 12 pct nickel, and 55 to 65 pct iron, with lesser amounts of molybdenum and manganese. The alloy is essentially free of zinc, lead, and sulfur. The zinc-lead fume obtained from the treatment contains about 40 pct zinc and 20 pct lead and represents essentially complete recovery of these metals.

The Bureau of Mines devised a direct flotation method for recovering potash from carnallite (KC1?MgC12?6H20) ore characterized by a high content of water-insoluble impurities. The method includes (1) a decomposition leach of the carnallite, (2) flocculation-depression of the insoluble slimes, (3) potash rougher, cleaner, and recleaner flotation, and (4) final product leaching. The technique eliminates the necessity for removing the water-insoluble slimes by mechanical or flotation de-sliming. Laboratory results demonstrated that the direct flotation procedure, when compared with conventional techniques, yielded equivalent or improved flotation results with equal or less reagent consumption. Locked-cycle testing of the direct flotation method recovered 87.8 pct of the potassium in the flotation feed in a final product containing 60.6 pct K20. A flowsheet incorporating the carnallite leaching-potash flotation procedure is presented.

This Bureau of Mines report describes laboratory and field experimentation in building mine ventilation stoppings, air-door frames, and fan bulkheads with a civil construction technique. Shotcrete is applied to a free-standing, three-dimensional wire-truss panel, which is easily cut and rapidly erected. In mines where shotcrete equipment is available, this technique should be faster and cheaper than conventional block-and-mortar or monolithic concrete construction.

During the latter part of 1943, a magnetometric survey was conducted by the Geophysical Division of the Bureau of Mines at the Mahopac magnetite mine, Putnam County, N. Y., in an endeavor to locate a segment of the ore body that reports state was cut off by a fault. The Mahopac mine was abandoned about 1895 after producing nearly 1,000,000 tons of high-grade magnetite iron ore. The results of the investigation indicate the termination of the ore body by "pinching", or a displacement of such magnitude as to place any extension at a considerable distance from the original are body. This report describes the work done on the project and presents the data obtained.

A close look at the Loyalhanna Limestone of southwestern Pennsylvania reveals a complex structural environment. Most exposures of the Loyalhanna occur along Chestnut Ridge and LaurelHill within the Allegheny Mountain Section of the Appalachian Plateaus Province. Because the Loyalhanna is rated as a super-pavement aggregate by the Pennsylvania Department of Transportation and because of its proximity to the population centers of southwestern Pennsylvania, the Loyalhanna has been extensively mined along these prominent anticlinal structures. Geologic and engineering analyses were performed using gas well and core logs, outcrop examinations, underground observations, and mine maps. Strata exposures from both outcrops and quarries were used to construct geologic maps, stereo net, and rose diagram projections so that the Loyalhanna’s structural environment could be understood better. Many of these structural conditions cause ground control problems at local quarries. These problems ranged from small-to-moderate sized rock falls associated with roof jointing to pillar failures associated with dipping discontinuities. Stresses ranged from tensional on structural domes where weathering dominates, to high levels of compression within structural saddles. A greater understanding of these characteristics is a prerequisite to develop engineering controls, such as improved mine layouts, pillar sizes, etc., that lessen miner exposure to these hazards.

Researchers at the Bureau of Mines have developed a technique for re- generating chromic acid-sulfuric acid etching solutions used in metal surface treatment operations. The technique utilizes a diaphragm cell equipped with a cation-selective membrane to oxidize cr3+ to cr6+ at the anode and to remove copper, the major metallic contaminant, at the cathode. Normally, spent etchant is discarded after approximately 3 days of use. Using the electrolytic cell, the etchant can be used for a year without replacement, From data obtained from industrial-scale cells, the installation of a regeneration cell with a 1,000-gal catholyte- holding tank has been estimated to save at least $240 per day, The payback period for the investment is estimated to be about 10 mo or less. Because the magnitude of these cost savings will vary at different locations, several graphs are presented to aid in calculating payback for a specific site. Using these graphs and the capital costs presented in this study, the payback period can be determined for installing a regeneration cell with a 500- or a 1,000-gal catholyte-holding tank in an existing surface treatment plant.

Two examples of en echelon mining-induced fractures seen in hard-rock mines provided a basis for inferring that fracture zones and bedding plane separations immediately surrounding mine openings are promoted by oblique shear into the openings. It is hypothesized that initial fractures or separations form at the corners of openings as a result of high stress and physical constraint on the rock's ability to deform elastically toward the opening. These conditions result in a locally preferred direction of shearing. The shearing, in turn, generates tensile stress that initiates a progression of systematically offset fractures approximately parallel to the direction of greatest compressive stress. The fractures or bedding separations create tabular rock layers that amplify shearing displacement through bending and dilation. Such shearing effectively reduces and redistributes the compressive stress, but significant dilation is an inevitable consequence. The combination of dilation and shearing and the progressive development of fracture zones have important implications with respect to ground support. The concept of mining-induced fractures forming as a result of shear is illustrated by two examples from coal mines. First, fractures seen at longwall faces probably result from shear associated with subsidence. The fracture zone that develops approximates or possibly defines the draw angle of subsidence. As the face advances, fractures extend downward along the lower edge of the fracture zone, while upper extensions of the fractures are pressed closed. Fracture zones in entry roofs provide a second practical example. Here, mining-induced fractures typically follow bedding planes. The shear zone model suggests that the first bedding separations develop near the edges of the roof and successive separations progress upward and toward the center. However, if the direction of greatest stress is inclined with respect to the roof, a fracture or bedding separation zone may propagate from one side only and also extend higher. Because coal ahead of the face provides some support against lateral shear deformation, bedding separation is inhibited near the face. Rock bolts installed close to the face ultimately become more strained and bent than bolts installed a few meters from the face, and bolts installed through the more remote part of a separation zone may ultimately experience the greatest tensile and bending strains. This model is supported by field data documenting progressive bolt failures that rapidly propagated downward across the roof during face advance.

NIOSH’s Pittsburgh Research Laboratory is currently involved in research to identify silica dust generation in underground metal/nonmetal mines. The ultimate goal of this research is to develop control technologies to reduce worker exposure to respirable silica dust. Commodities and job classifications with the highest silica dust exposure have been identified through analysis of the Mine Safety and Health Administration (MSHA) compliance dust sampling database. Dust surveys were conducted in an underground limestone and gold mine to investigate silica dust sources, generation levels, and controls being used. A summary of the mining operations, sampling procedures, resulting data, and suggested methods to improve the dust control will be provided.
