"Effects of Bisbenzylisoquinoline Alkaloids on Alveolar Macrophages: Correlation between Binding Affinity. Inhibitory Potency and Antifibrotic Potential. CASTRANOVA, V., KANG, J-H., MA. J. K. The Chinese have conducted extensive studies concerning the medicinal properties of plant products. In this investigation the ability of three bisbenzylisoquinoline alkaloids to inhibit particle-induced activation of alveolar macrophages was evaluated and this inhibitory potential was correlated with the ability of those drugs to bind to membrane components. Tetrandrine. i.e.. an herbal medicine used as an antifibrotic agent in China, was a potent inhibitor of particle-stimulated oxygen consumption, superoxide release, and hydrogen peroxide secretion by alveolar macrophages. Tetrandrine also exhibited substantial binding affinity for membrane lipids and alveolar macrophages. In contrast, tubocurine, an analogue with little antifibrotic potential, exhibited low binding affinity and had little effect on macrophage activation. Methoxyadiantifoline, an alkaloid of unknown antifibrotic potential, exhibited inhibitory and binding properties similar to those of tetrandrine. The data indicate that a strong relationship exists between the antifibrotic potential of these alkaloids and their ability to bind to alveolar macrophages and inhibit particle-induced activation of these phagocytes. These drugs should serve as useful probes to evaluate the role of alveolar macrophages in pulmonary fibrosis. © 1991 Academic Press, Inc.Chronic silicosis is a fibrotic disease resulting from occupational exposure to silica dust. It is manifested initially by an increase in the quantity of pulmonary surfactant (Dethloff et al., 1986). However, in later stages it is characterized by an increase in collagen formation and the development of concentric hyalinized nodular lesions in the lung (Ziskind et al., 1976). Although mechanisms for the progression of silicosis have not yet been completely defined, activation of alveolar macrophages by respirable silica particles is believed to play an important role in the etiology of this disease. Studies from our laboratory have shown that both in vitro and in vivo silica exposures result in a stimulation of macrophage release of reactive products capable of damaging the lung parenchyma (Castranova et al., 1990; Goodman et al., 1990). In addition, exposure of al¬veolar macrophages to silica results in the release of fibrogenic factors which enhance the proliferation of fibroblasts and the synthesis of collagen by these pneumocytes (Leibovich and Ross, 1976; Bitterman et al., 1982, 1983).The bisbenzylisoquinoline alkaloids, which represent the largest group among the iso-quinoline compounds isolated from the plant kingdom, are known to exhibit a broad rangeof pharmacological activities. Tetrandrine, for example, an alkaloid isolated from the root of Stephania tetrandra, has been tested for its antitumor, antihypertensive, and antisilicotic effects (Kupchan and Atland, 1973; Zeng et al., 1985; Yu et al., 1983). It is currently used in China as a treatment for angina, hypertension, and silica-induced pulmonary fibrosis (Li et al., 1981). Several related bisbenzylisoquinoline alkaloids such as methoxyadiantifoline and tubocurine have also been evaluated for medicinal effects. Methoxyadiantifoline has been shown to exhibit effects on the cardiovascular system similar to those reported for tetrandrine (Jia et al., 1988). However, to date, the antifibrotic activity of methoxyadiantifoline has not been evaluated. In contrast to tetrandrine, tubocurine, a structural analogue of this drug, has been reported to be relatively ineffective against fibrosis (Mo et al., 1982)."

The Bureau of Mines estimated the potential availability of iron ore from 129 domestic and foreign mines and deposits in 25 market economy countries. The availability of four products on a total and regional basis was determined from demonstrated resources of 75.3 billion It. From the resources evaluated there are a total of 18.6 billion It of sinter fines, 4.9 billion It of lump ore, 14.7 billion It of pellets, and 820 MMlt of pellet feed potentially available at a 15-pct DCFROR, f.o.b. port. On a regional basis, at or below a total cost of $0.26 per iron unit there are 1.7 billion It, 2.8 billion It, and 186 MMlt of sinter fines potentially available from Brazil, Australia, and Africa, respectively. There are 825 MMlt of lump ore from Brazil and 1.4 billion It from Australia potentially available at or below a total cost of $0.24 per iron unit. There are 2.1 billion It of pellets potentially available from the United States at or below a total cost of $0.80 per iron unit, while 442 MMlt are potentially available in Brazil at or below $0.38 per iron unit. South America has 208 MMlt of pellet feed potentially available at or below a total cost of $0.28 per iron unit. While the resources of iron ore are more than adequate to satisfy demand, the future of the iron ore industry is very dependent upon changes in the world economy, and on the domestic scene, changes in the domestic steel industry and steel imports.

This U.S. Bureau of Mines report reviews the state of the art in blind pneumatic stowing of backfill in voids in abandoned coal mines. It contains a short history of the development of pneumatic equipment in mining and four recent case histories of blind pneumatic stowing. Limitations of pneumatic stowing in terms of equipment wear, costs, backfill material, and methodology are discussed, and some new procedures and designs are reviewed. A bibliography covering pneumatics in mining is included also. This report provides background for the preparation of a Bureau test program in which the variables affecting the efficiency of pneumatic backfilling will be studied using a full-scale surface test facility.

"INTRODUCTION As part of its work in promoting safety, the Bureau of Mines is investigating various means of eliminating or mitigating the explosion hazards of combustible gases encountered in mining and industry. Information has been obtained and a procedure developed whereby the explosibility of many complex gaseous mixtures may be calculated in a rather simple manner without the necessity of performing laborious laboratory experiments. An earlier publication 3/ described the method of determining by calculation the limits of inflammability of gaseous mixtures, including methane, hydrogen, carbon monoxide, nitrogen, carbon dioxide, and oxygen. The information published in the above report permitted calculation of the limits of inflamnability of such gases as are liberated by mine fires, gases from explosives, automobile exhaust gases, gases present after mine explosions, blast-furnace gas, and other gases of similar type.Gases liberated in certain potash mines in the western part of the United States sometimes contain ethane, in addition to the gases named. An investigation was made to obtain data from which the limits of inflammability of such gaseous mixtures could be determined by calculation. 4/Similar data have been Published covering all types of natural gases containing wide ranges of concentration of nitrogen and carbon dioxide.5/"

The storage stability characteristics of a diesel type fuel were determined to evaluate the effect of the polar compounds in the fuel. These compounds, comprising most of the nitrogen and oxygen containing compounds and many sulfur containing compounds, were separated from the fuel with a chromatographic column. This fraction was further separated into three polar extracts by graded elution through the column. Each extract was characterized. A storage program using the depolarized fuel, the fuel as received, and blends of the extracts with both fuels, was conducted. Results showed that the removal of the more polar components stabilized the fuel. Blending the polar extracts with the original fuel and with the depolarized fuel in the same ratio as they existed originally and at two and three times the original ratio increased the amount of gum present before storage, but had little effect on the formation of additional gum during storage. Distillate fuels produced by commercial refining of petroleum contain traces of organic nitrogen, sulfur, and oxygen compounds that are more polar4 than the remainder of the fuel. Recent laboratory studies by the Federal Bureau of Mines (1),6 in which these compounds were removed from a fuel, have shown that they influence its storage stability. The objective of this work was to determine the amount of these compounds present in a fuel and their effect upon its stability in storage. To evaluate the role of these polar compounds, it is necessary to remove them from the distillate fuel. Many can be removed by chemical treatment (2), but such treatment precludes reblending the polar components to approximate the original fuel composition. Solvent extraction is free of some of the criticism of chemical treatment, but it is more difficult because of the complex mixture from which compounds must be removed.

This chapter presents an overview of fatal and nonfatal injuries reported to MSHA during 1986-1995. Reportable injuries follow the criteria for reporting specified in 30 CFR 50. These criteria are listed in appendix A. The data used for these analyses came from the MSHA accident, injury, and illness data closeout files for the years 1986-1995. The MSHA data are described in appendix A and methods used in data analysis in appendix B. There may be slight discrepancies between the figures presented here and other presentations of MSHA data for the following reasons: (1) injuries to nonemployees have been excluded from analysis; and (2) injuries identified by MSHA after the files were closed out are not included. Within the MSHA data, there are differences in reporting requirements for operators and contractors. Contractors are required to report only those injuries that occur during certain categories of work (described in appendix A). For this reason, data on injuries among employees of operators and contractors in the MSHA database should not be considered directly comparable. As discussed in the "Introduction," the MSHA data on fatal injuries differ from the CFOI data (chapter 1) because of varying definitions used for the mining industry. As compared to the CFOI data, MSHA data exclude injuries that occurred off of mine property and include injuries occurring at mills and processing plants on mine property. Tables 4-1 through 4-9 and figure 4-1 provide comparisons of injury frequencies and rates among the commodities. These comparisons are followed by sections 4A through 4E, which provide more detailed tables and figures for each commodity separately. The method used to calculate injury rates is described in appendix B. Since denominator data for occupations (lob titles) were available for 1986 only, estimates of rates by occupation for the entire 10 year period are based on extrapolation from 1986 data. For this reason, estimates of injury by occupation should be interpreted with caution. Fatal Injuries A total of 1,078 fatal on-the-job injuries were reported to MSHA during the 10-year period 1986- 1995. The average annual fatal injury rate for all commodities combined during this period was 30.9 per 100,000 full-time equivalent workers. Mine operators accounted for 908 (84%) of the fatalities and had an average annual fatal injury rate of 28.2 per 100,000. While mine contractors comprised only 16% (n = 170) of the total number of fatalities, their average annual fatal injury rate was 2.3 times higher than that of mine operators (65.0 per 100,000). Table 4-1 presents the number and rates of fatal injuries by commodity for operators and contractors. Contractors in metal, nonmetal, stone, and sand and gravel are grouped together, since MSHA data on employment were not available for rate calculations for these commodities separately. The highest average annual fatality rate for the period, 66.5 per 100,000, occurred among metal and nonmetallic mineral mining contractors, followed closely by coal contractors (63.1 per 100,000). Among mine operators, the highest average annual fatal injury rate was in coal (38.9 per 100,000); the lowest was in nonmetal (14.3 per 100,000). Table 4-2 shows the average annual fatal injury rate by type of operation for each commodity for mine operators only. Of commodities with underground mines, all except stone had higher fatality rates at underground than at surface operations of these mines. The highest fatality rate for any operation occurred in surface operations at underground stone mines (124.5 fatal injuries per 100,000 workers). Stone had the second highest fatality rate for underground operations at underground mines (77.9 per 100,000). The highest rate for underground operations at underground mines occurred in metal (87.6 per 100,000). Table 4-3 shows the number and rate of fatal injuries by U.S. Bureau of the Census occupational groups (described in appendix D) for the 10 occupation-commodity groups with the highest fatal injury rates. These data should be interpreted with caution because the denominator used in calculating the rates was based on an extrapolation from data for the year 1986 only, as described in appendix B. Extractive occupations are represented in this group for three commodities: coal, metal, and sand and gravel. Construction trades are included for coal, sand and gravel, and nonmetal. The distribution of fatal injuries by commodity and work activity for mine operators is presented in table 4-4. A description of the work activity categories is in appendix C. Overall, the leading work activity at the time of injury was using or operating tools and machinery (26.4%) followed by constructing, repairing, and cleaning (24.0%), vehicular operations (18.8%), and materials handling (12.6%). The distribution varied by commodity. The largest percentage of fatal injuries occurred while operating tools and machinery in both coal and stone; during vehicular operations in metal; and during constructing, repairing, and cleaning activities in nonmetal and sand and gravel. Table 4-5 shows the distribution of fatal injuries by commodity and MSHA accident classification for mine operators. A description of the MSHA accident classifications appears in appendix E. The largest proportion of injuries overall occurred during powered haulage (28.5%). The

"INTRODUCTION A report of tests of seven small pillars formed from the Pittsburgh coal bed in the Experimental coal mine was given in Technical Paper 605 5/ Five additional pillars were tested between July 1939 and. October 1940. The data obtained, in the later tests permit an extension of the conclusions reached in Technical Paper 605, which appears to be important enough to the mining industry to justify a progress report that will explain briefly what it is and how it was obtained. Full detail of the tests will appear in a second technical paper, preparation of which is deferred so that results of pillar tests now under way my be included.DESCRIPTION OF TESTSExcept for minor improvements in operating details, the five additional tests were made as described in Technical Paper 605, pages 2 to 7. Briefly, the desired pillar was formed from the coal bed, by excavating a passage around it (no explosives were used), and enough material was removed from above to permit installation of the assembly used for applying pressure. Each pillar was tested resting on its original fireclay floor, which was restrained from lateral flow by a ring of concrete. Vertical and lateral movement of the pillars in two directions as measured carefully in all tests."

An essentially chemical technique has been developed on a small scale for the efficient utilization of certain domestic titaniferous iron ores. Three main steps are involved: (1) Sintering the ore at 1,050° C. with carbon and soda ash to reduce the iron to metal powder, retaining the titania in the slag, (2) separating the powdered iron from the slag, after wet grinding, by magnetic or gravity methods, and (3) sulfuric-acid decomposition of the titania fraction to produce pigment and certain byproducts. The technique was first applied to the tailings from the Bureau of Mines bauxite-concentrating plant at Bauxite, Ark. A good grade of powdered iron was produced from this material. Subsequently, the work was diverted to the study of the titaniferous magnetite from the MacIntyre Mine at Tahawus, N.Y., because of the potentially higher recoveries of iron, titanium and vanadium. Ninety percent of the iron in this ore has been recovered as an iron powder analyzing 90 percent metallic iron and containing 2 to 3 percent TiO2. The remaining titania was found in the slag and was recovered as pigment-grade TiO2 by application of a suitable technique. The vanadium was distributed between the TiO2 fraction and the alkaline liquor resulting from the wet-grinding and magnetic-separation operations, with only a minor proportion retained by the metallic iron.

On 26 August 1971, the Bureau of Mines, through the Spokane Mining Research Center at Spokane, Washington, approved initiation of the evaluation of oscillatory energy toward enhancing flow of polymer into shale. The effort was an outgrowth of work performed under Bureau of Mines Contract H0210055, "Feasibility Study of Surface Impregnation equipment for the Chemical Stabilization of Coal Mine Structures," wherein feasibility of the impregnation concept was expressed and the detailed design of a machine for impregnation of coal mine roofs with polymer (under static pressure head) accomplished. Under these efforts, the potential enhance¬ment of polymer uptake into the shale thru superposition of oscillatory energy on the static impregnation pressure was suggested. A proposal for investigation of this phenomena entitled "Feasibility Study of the Application of Oscillatory Energy as an Aid to Afford Chemical Stabilization" was presented to the Bureau whereupon a three month program for initiation of investigation was approved.

During World War II the shortage of ore revived interest in eastern iron deposits, among which -!2..c "Ile Massachusetts and Connecticut limonites. The Cheever mine, in the Richmond-Salisbury area of Massachusetts, was formerly a large producer of limonite ere. The Bureau of Mines investigated the lateral and downward extensions of the Cheever ore body, which was mined from about 1850 to 1892, by ap open cut 1,000 feet long and also by underground workings. even holes aggregating 1,723 feet, were drilled by the Bureau during the winter of 1943-44. They comprised three diamond-core drill holes, two churn-drill holes, and two rotary-drill holes. A geophysical survey of the Cheever mine area and adjoining ground was made in conjunction with the drilling program. Although this survey did not locate the limonite ore beds, the data were of considerable assistance in locating the limestone contact which forms the footwall of the ore beds. Drilling disclosed two beds of limonite with a heavy overburden of glacial till.

Diesel-powered haulage equipment has recently been tested and approved for permissibility by the Bureau of Mines for use in mining in the United States. Achievement of this advance in safety involved extensive studies, both in the testing station (2, 3, 20, 21, 22, 23, 24, 25, 26, 27)3/ and in the field 1, 4, 13, 30), of the operating characteristics of diesel engines in relation to the toe of such engines underground. The results of these studies formed the basis for establishing the requirements that should be met by diesel-powered haulage equipment approved for permissibility. Considerable interest has been manifested in the development of diesel-powered haulage equipment fur mines. The purpose of this paper is to retrace briefly the course of this development and to answer such questions as: What is the history of the use of diesel engines underground? Why has the Bureau of Mines been interested in the development of diesel-powered haulage equipment for use in mines?

Prier to the date of this contract, Ocean Energy had developed and demonstrated a high efficiency fluorescent caplamp which operated from a standard 4 volt miner's caplamp battery. This unit produced about five times the light output of the standard incandescent miner's caplamp. The Company believed that the major market for this unique caplamp was in the industrial and municipal applications. Because of the additional safety requirements for coal mining use, and in conjunction with the much smaller market in the mining industry, the additional expense and difficulty of modifying this caplamp to comply with the Federal and State coal mine regulations were not considered at that time. Recognizing the potential of this lamp for improving illumination conditions for underground coal mining, the US Bureau of Mines contracted with Ocean Energy to make the required modifications to this lamp to bring it into compliance with the Federal and State of Pennsylvania regulations, and thereby, make this lamp available to the anthracite and bituminous coal mining industry. At the outset of the contract, the injection molds for the caplamp housing were being completed, and only prototype, hand fabricated, units were available for tests. The Bureau evaluated these prototype units and formulated a performance specification for the miner's version. Light level and distribution, weight, size, angle of tilt, and operating duration were all specified. In addition, it was required that the caplamp be com¬patible with existin charging equipment at the mines and that the final design be in full compliance with MESA Schedule 6D and he ultimately approved by MESA, Approval and Testing, as well as by the Pennsylvania Bureau of Deep Mine Safety. The scope of this contract covered the modifications required to achieve these objectives. It was agreed by the Bureau that all rights to the lamp, including design, material combinations, circuitry, etc. would remain with Ocean Energy. The charger/adapter system and components, which were developed under contract funds, are disclosed hero and become public knowledge and property. Patent applications have been filed by Ocean Energy to protect the mechanical and electrical design aspects of the lamp. It should be noted that this report contains details, designs and data which may be covered by one or more Ocean Energy patent applications, and disclosure herein is made only to provide better understanding of the contract results.

This is one of a series of reports being published by the Bureau of Mines on mining methods and costs at various mines throughout the United States. Including the Holden mine in this series is particularly appropriate at this time. The mine was operated on a low-cost, large-scale production basis to compensate for low-grade ore. Even with the best conditions and the most efficient methods, the margin of profit was small. Under the present situation of rising costs and lowering metal prices, the profit margin vanished. On June 28, 1957, the mine operations ceased; therefore, this report can review the entire life span of the mine.

The underground junction or intersection is analogous to the doorway or window in a building in that the continuous wall or support for the overhead structure has been weakened by making the opening. Some type of bridge or support seems necessary to prevent roof sag and possibly eventual failure at this point, although frequently it appears that little is done until a fatal accident induces action. The intersection generally represents less than 15 percent of the exposed roof area. As a result of recent multiple fatalities, some of which occurred at or near intersections, the Bureau of Mines was asked to examine the junction-support problem more thoroughly and to report its findings. A national survey of the junction-support systems in use has been made by correspondence, and the local area has been canvassed rather thoroughly to determine just what and how much is being done.

West Virginia ranks second in the nation in coal production, surpassed only by Wyoming, and usually accounts for 15 to 20percent of the nation's total annual production. Cod occurs in seams of minable thickness in all but nine of the state's 55 counties. There are two coal- producing regions, the northern field and the southern field. West Virginia coal is mined by both underground and surface methods. Underground mines in West Virginia account for a major portion of its coal production. Surface mines are generally contour operations and, especially in the southern field, are in steep slope areas, with slopes greater than 20 degrees. Many of the surface mines remove multiple seams. Recoverable reserves in West Virginia total in excess of 55 billion tons of bituminous coal. West Virginia's regulatory program was conditionally approved on January 19, 198 1. Its Abandoned Mine Land Program was approved February 23,198 1, with emergency authority granted on August 26,1988.

This Bureau of Mines report compares the availability of federally owned minerals in Arizona with lands considered favorable for discovery of mineral resources, to deter-mine the extent to which Federal mineral lands in such areas have been withdrawn by legal action or restricted by management practice. Of particular concern are restraints on Federal lands in areas identified as highly favorable for occurrences of valuable mineral deposits. Excluding Indian lands held in trust by the Federal Government, about 32 million acres, 44%, of Arizona's mineral estate is federally owned. This total includes lands with partial Federal mineral ownership. Approximately 16.5 million acres, or 52% of these lands, are available for location or lease. This includes 496,000 acres, 62%, of the lands highly favorable, and 2.6 million acres, 57%, of the lands moderately favorable for locatable minerals in Arizona. All other Federal mineral lands are either withdrawn or restricted, reflecting a wide range of purpose, authority, and impacts on mineral exploration and development.

The Bureau of Mines laboratory tested the response of a commercially available real-time aerosol monitor (GCA RAM-1) to various dusts. Monitor measurements were recorded, averaged, and compared with simultaneous gravimtric measurements of each test dust. Tests usually lasted several hours. The test dusts of various particle size distributions used included coal, limestone, and a commercially available test dust. For each particular dust, the monitor response was linear and correlated well with mass concentration over the range of about 0.5 to 10 mg/m3. The monitor can estimate a 2.0-/m3 respirable coal dust concentration within as little as 26 pct with 95 pct confidence. The monitor must, however, be calibrated with the dust to be measured because the instrument response is affected by the type of dust particle. The average monitor response to same mass concentration of coal dust was approximately twice the average monitor response to the same mass concentration of limestone dust.

Two-stage electric smelting tests, using small submerged arc furnaces, were made on high-iron manganiferous materials from the Pioche District, Nev., and the Cuyuna Range, Minn. The object was to reduce sufficient iron in the first stage to give a slag with a favorable Mn to Fe ratio for smelting to ferromanganese in the second stage. The nodulized flotation concentrate from the Pioche District contained 29.2 percent Mn and had a Mn to Fe ratio of 2.3:l. When smelted with coke and silica or alumina as flux, more than 90 percent of the manganese was recovered in slags with Mn to Fe ratios of 10: :24:1. The Pioche slag was smelted to ferromanganese using coke and lime- stone or dolomite as flux. The ferromanganese was high in silicon but other- wise met specifications. Recovery of manganese to ferromanganese in this step was 73 to 78 percent. Overall recovery of manganese was about 70 percent. The sintered flotation concentrate from the Cuyuna Range material contained 12.9 percent Mn and had a Mn to Fe ratio of 0.3:1. When smelted with limestone as flux, about 83 percent of the manganese was recovered in a slag with a Mn to Fe ratio of about 6:1. The Cuyuna slag was smelted to ferromanganese with coke and limestone for flux. Recovery in this step was 61 percent of the manganese in a high silicon ferromanganese. Overall recovery of manganese was about 50 percent.

Case 1: A driller raised a drill rig mast into a 7200-volt overhead power line. Both he and his helper were killed. Case 2: A driller-helper was pulling a winch cable toward a stack of drill steel when the cable contacted a 12,000-volt power line. The helper was killed, and the driller at the rig controls was severely injured. [ ]
