The mining industry has the highest average annual fatality rate among major U.S. industries. To address this, the National Institute for Occupational Safety and Health, Pittsburgh Research Laboratory (PRL), is conducting major research programs to reduce the hazard exposure of miners. One of the recently concluded programs at PRL developed the enabling technology for remote-controlled mining. An application involves the manual process of extracting and hauling coal where operators, in the tight confines of a mine, can be struck or caught by mobile machinery. The approach to remedy this problem uses a guidance system on the continuous haulage equipment so that it follows the continuous mining machine that extracts coal. This, in essence, involves sensor-based docking of the machines. Sensors functioning in the hostile mine environment of dust, methane gas, and water play the key role. Computer analysis of the mining machine's movements and empirical machine characterizations established operating requirements and spatial limitations to ensure proper loading of coal into the haulage equipment. These data served in the selection of a guidance sensor. Technologies such as scanning laser systems and ultrasonic sensors have frequently been used in other applications, but were found unacceptable. However, a near-infrared sensor employing active targets met the requirements. The sensor has a nominal 75b conical field of view and a range from 0.1 to 18.0 m. For the single-target mode, nominal range accuracy was 4.3% at a distance of 3.65 m. Correction algorithms were generated, which reduced the error to 0.6%. Airborne dust testing showed less than an additional0.8% accuracy (worst case) degradation at dust levels up to 15 mg/m3. Using four targets, the nominal range accuracy was 0.4% without correction algorithms. Analysis of dynamic testing of a continuous miner and haulage showed that accuracy was maintained and total target loss did not occur. A guidance system for the haulage system to follow the mining machine does not exist commercially. Such a system can reduce fatalities, injuries, and exposure to dust and noise.

Laboratory crushing experiments were conducted on a range of low- to high-volatile bituminous coals to investigate the various factors influencing airborne respirable dust (ARD) generation. This research was conducted to identify the principles of ARD liberation from the coal product. Five U.S. bituminous coals were uniformly prepared and processed through a double roll crusher located in a low-velocity wind tunnel. Experimental factors studied included inherent coal seam constituents, coal grind ability, specific energy of crushing, product size characteristics, dust cloud electrostatic field, and specific ARD generated. The results of this investigation indicate that a combination of several factors are associated with ARD generation. One factor is the effect of coal rank, described by the inherent moist fuel ratio, on the product size characteristics, defined by Schuhmann size function parameters. Another key factor is the effect of air dry loss(ADL) moisture in the coal seam on the breakage-induced electrostatic field of airborne dust. The effect of these factors is that different percentages of <10-m coal particles are dispersed as ARD. A discussion of electrostatic field principles, coal ADL, and its effect on ARD generation is presented.

The behavior of wire ropes used in hoisting is not well understood. In an effort to improve this understanding, the structure of round-strand wire ropes was analyzed. This report provides a generalized mathematical model that completely describes the geometry of the wires. It consists of two sets of vector equations and is valid for any round-strand wire rope. One set of equations is used to trace the paths of wires that have the form of a single helix; the other is used for the paths of double helical wires. The specific model for a 33-mm 6×19 Seale, independent wire rope core (IWRC), right regular lay wire rope was presented as an example. The paths and the geometric properties of the wires, which include the path length per lay of strand, the curvature, and the torsion, were determined from this model. The effects of strain deformation were analyzed, resulting in a system of equations for determining the structural parameters of the deformed rope. In future work, the model could be used to analyze wire ropes of different constructions so that a more scientifically based understanding of rope behavior can be established.

Coal dust and float coal dust, produced during normal mining operations, in underground coal mines, are carried from the point of origin downstream by the ventilating air, where it deposits on the surfaces of the mine entry. In an explosion, this dust is lifted from the surfaces by the aerodynamic disturbances and, if of sufficient quantity, can continue to propagate the explosion. To prevent the surface coal dust from contributing, it must be inerted, typically by spreading pulverized limestone, i.e., rock dust, over the coal dust surface. To facilitate the dusting operation, the National Institute for Occupational Safety and Health (NIOSH), Pittsburgh Research Laboratory(PRL), developed an automated system that continuously monitors the accumulation of coal dust. This system could activate a rock-dusting machine that disperses rock dust into the ventilation air when dangerous deposits accumulate and deactivate the machine when sufficient inert has been deposited on top of the coal dust. The system consists of a microprocessor-controlled optical float dust deposition meter. This device measures the light intensity reflected from a deposited layer of dust. A standard cap lamp is used as a fixed-position light source. From the reflected light signal, the microprocessor determines the hazard level of the deposited layer and performs the appropriate action. Full-scale studies conducted in the Experimental Mine at PRL&apos;s Lake Lynn Laboratory, using alternating thin layers of rock dust and coal dust, successfully demonstrated the operation of the device. Based on the statistical data from these studies, a mathematical model was developed to predict long-term error in total rock dust content using this inerting procedure. Downwind dust dispersion was also examined to provide optimum placement of the dust sensors and to determine the need for better rock dust dispersion techniques.

"Inhaled dust particles of appropriate size contact cells of the alveolar region.' The acute response to coal dust appears to involve a variety of cell types, including alveolar epithelial cells, interstitial fibroblasts, both resident and recruited macrophages, and cells of the immune system. Direct and indirect effects of dust, which may be mediated by soluble factors,'' can initiate acute and long-term lung injury. While it appears that epithelial cell growth and differentiation, along with integrity of the epithelial basement membrane, may play a role in the long-term response to dust exposure, little is known of the role the alveolar epithelium may play in the progression of injury. The broad hypothesis that underlies these experiments is that short- and long-term consequences of coal dust exposure involve changes in structure and function of the alveolar epithelium and the underlying extracellular matrix. Recent studies show that coal dusts exert direct effects to modify pulmonary epithelial cell-extracellular matrix interactions in vitro.3EXPERIMENTAL PROCEDURESType II cells isolated on day 0 by density centrifugation and differential attachment4 were plated overnight in D MEM containing 10% fetal calf serum before exposure to anthracite dust PSOC 867 sonicated into culture medium (750 ug/mL).3 Synthesis of extracellular matrix proteins was determined on day 3 of primary culture, as detailed elsewhere. 5 Cells were removed from extracellular matrix by extraction with NH4OH and high salt. Extracellular matrix synthesis and fibronectin content were evaluated based on radiolabeling with 3 H-leucine and on Western blot analysis, respectively. "Lee, Yu-Chen, and D. Eugene Rannels. Alveolar macrophages modulate the epithelial cell response to coal dust in vitro. Am. J. Physicl. 270 (Lung Cell. Mol. Physiol. 14): L123-L132, 1996.-The response of the alveolar epithelium to coal dust exposure is poorly understood. Coal or other dusts may act on the epithelium directly or indirectly through nearby alveolar macrophages (AM) that produce cytokines and other soluble products. AM and type II pneumocytes (T2P) were thus exposed to dust in coculture to evaluate their possible interactions. Anthracite coal dust PSOC 867 increased synthesis of extracellular matrix (ECM) components by T2P. AM alone did not produce ECM. Similarly, coculture ofT2P with AM (3.75:1) had little effect on epithelial ECM synthesis. In contrast, coculture ofT2P with AM significantly increased PSOC 867 effects on T2P rates of ECM synthesis, ECM fibronectin content, and T2P levels of fibronectin mRNA. AM-conditioned medium did not change the PSOC 867 effect on T2P. Neither control nor PSOC 867-treated AM on Falcon culture inserts (0.45-µm pore size) over T2P stimulated ECM synthesis by either untreated or dust-exposed epithelium. Thus AM-mediated changes in ECM synthesis by PSOC 867-treated T2P require close cell-cell interactions, suggesting a role for cell-cell contact or for short-lived soluble mediators of the AM effects. DEVELOPMENT OF PNEUMOCONIOSIS, a chronic disease of coal workers that results in pulmonary fibrosis and low lung compliance, is associated with inhalation of mineral or coal dust in the work place (14). Although the pathology of pneumoconiosis has been studied extensively, cellular mechanisms that cause changes in lung structure and function are not fully understood. Particulate matter that enters the respiratory system distributes according to size, with larger particles being removed in proximal airways. Dust components of small size may enter distal alveolar region, which is populated by two types of epithelial cells, type I and - type II cells, and by alveolar macrophages (16). Each of these cell types may interact with inhaled particles, but only the effects of coal dust on alveolar macrophages have been studied extensively. Less is known regarding the response of the alveolar epithelium, or of effects on epithelial cell-alveolar macrophage interactions, in the dust-exposed lung.Function of the alveolar epithelium is well known to be influenced by the underlying basement membrane, or extracellular matrix (28). Alveolar epithelial cell extracellular matrix interactions, which may involve modulation of cell migration, attachment, or differentiation (17), are reflected in type II cell morphology, metabolism, secretory activity, hormone responsiveness, lamellar body content, and gene expression (21). The extent to which basement membrane modulates differentiation of the overlying epithelium is determined by both molecular composition and three-dimensional structure of the matrix. Type II epithelial cells in vitro synthesize, secrete, assemble, and degrade a variety of basement membrane components, including laminin, fibronectin, type IV collagen, and thrombospondin (22, 27). How expression of these components is regulated, directly or indirectly, upon exposure of the alveolar epithelium to injury by coal dust or by other particulates remains to be resolved."

"The evolution during oxidation of the internal structure of soot and carbon black particles was studied using high-resolution transmission electron microscopy (HRTEM) and an image analysis system. Increasing ordering to the carbon structure with increasing oxidation is observed. Measures of the increased order are increases in the fractional coverage of a cross section of the particles with a layered structure, a decrease in the mean interlayer spacing, and a decrease in the spread of the interlayer spacing. The changing structure of the carbon black impacts its properties, such as the rate of oxidation.There is considerable interest in the study of the structural changes of carbonaceous materials as a function of oxidation. Davis et al.1 measured the variation of carbon dioxide surface area of various coals as a function of residence time in a flow reactor. HRTEM measurements revealed that crystallite dimensions and interlayer spacings are altered by combustion. Ishiguro et al.2 showed that oxidation alters the structure of diesel soot. The aim of our current research is to develop a better understanding of the microstructural reordering of carbon blacks with implications to their properties. The carbon blacks used in this research have already been studied and their reactivities as a function of oxidation measured.3The methodology used in this work has already been described in detail elsewhere.4 This paper presents an application to quantify the structural changes of carbon black particles as a function of oxidation and graphitization. The technique used is based on the use of HRTEM (high-resolution transmission electron microscopy) and computational image analysis.It is well-known that carbonaceous materials like graphite, soots, coals, chars, etc., possess characteristic structural appearances which can vary from mostly random or amorphous to a perfectly ordered graphitic crystalline structure. The carbon structure has been variously described as turbostratic5 or as crumpled sheets6 to indicate that parallel layering, reminiscent of graphite, is observed over dimensions of nanometers but that these planes or crystallites show disorder over larger scales. The degree of order of such structures is strongly dependent on the thermal treatment of the material as well as the composition of the source of the carbonaceous material. The value of electron micros¬copy in studying the morphology of such materials has been well established,2,7,8 although only a few attempts toward quantification have been made.The sample observed was carbon black CB330 from De¬gussa. One set of carbon black samples were oxidized in a thermo-gravimetric analyzer (TGA) in air at 575 0C, so that the conversion varied from 0 to 96%. Another set of carbon black samples were graphitized (Graphpac-GB by ALLTECH) and then oxidized in the TGA at 680 °C until 92% burnout was reached. The latter samples are called GB 0% and GB 92%, respectively.For the HRTEM observation a small portion of each sample was ultrasonically dispersed in ethanol. The suspension was deposited dropwise on a copper TEM grid coated with a lacey carbon film. The examination of the samples was carried out on areas that extend over the holes in the supporting film in order to avoid interference from the amorphous carbon background film An oriented gold crystal was used as a calibration standard and subjected to the same techniques as the carbon black samples."

In spite of advances in measurement techniques and an explosive increase in the volume of research, our understanding of human strength remains in its introductory stages. It is clear that muscle strength is a highly complex and variable function that depends on a large number of factors. It is not surprising, therefore, that large differences in strength exist not only between individuals, but even within the same individual tested repeatedly on a given piece of equipment. The issue is compounded by the fact that correlations of strength among different muscle groups in the same individual are generally low, and tests of isometric strength do not necessarily reflect the strength an individual might exhibit in a dynamic test. As a result of these and other influences, great care needs to be exercised in designing, evaluating, reporting, and interpreting muscular strength assessments. Traditionally, tests of muscular strength were in the domain of the orthopaedist, physical therapist, and exercise physiologist. Such tests are also an important tool for the ergonomist, however, because of the high strength demands on workers in manual materials-handling tasks. In some cases, task demands may approach or even exceed the strength that an individual is voluntarily willing to exert in a test of strength. In such cases, evidence suggests that the likelihood of injury is significantly greater than when the task demands lie well within an individual&apos;s strength capacity. Because the relationship among strength capabilities, job demands, and musculoskeletal injury has been established, it is apparent that tests of muscular strength may benefit the ergonomist both in designing jobs and in ensuring that individuals have sufficient strength to safely perform physically demanding jobs. Several strength assessment techniques have been employed for these purposes, each possessing unique characteristics and applicability to job design and worker selection procedures. Our main purpose has been to elucidate these strengths and weaknesses, so that tests of strength may be properly applied in designing jobs and selecting workers. One of the crucial points we have emphasized is that any test of strength used in job design or worker selection must be directly related to the demands of the job.(1) For example, if an occupational lifting task has a high dynamic component, a test of isometric strength is not likely to provide the data necessary for proper design of the job. Of course, dynamic strength tests would also be misapplied in assessing a job requiring isometric exertions. Another potential pitfall is using tests of strength on isolated muscle groups and assuming that they are indicative of whole-body strength. For example, one might mistakenly assume that dynamic trunk extension strength represents a person&apos;s capability to perform a lifting task. However, an individual&apos;s lifting capacity may be entirely unrelated to trunk extension strength. It may, instead, be limited by an individual&apos;s arm or leg strength, depending on the task being performed. A final point on strength assessment should be made. An individual&apos;s strength capability cannot be considered a fixed human attribute. Strength training regimens can increase an individual&apos;s strength capability by 30%-40%. Whether

Cable bolts are emerging as the newest ?twist? for roof support in U.S. underground coal mines. For decades cable bolts installed in underground metal mines in the United States and Canada used cement-based grouts for anchoring. The cement anchoring process, because of the time and expense for installation, made cable bolts impractical for use in coal mines. Today, the introduction of resin-anchored cable bolts provides a system more consistent with tradi­tional U.S. coal mine roof bolting practices and requirements. Expecta­tions are that the utilization of cable bolts for a number of U.S. mining applications will expand, particularly in coal mining.

? A roof bolter reaches for a lever while guiding the drill with his other hand. Since he&apos;s watching the drill and many of the controls look and feel alike, he hits the wrong control and crushes his hand. ? Six hours into the night shift, an ore truck driver dozes off. The truck plows over the berm and down into the pit. ? Fleeing a afire, a face crew sees smoke in the intake entry. Assuming the fire is ahead, they cross over to another entry, but soon encounter even worse smoke. These very different accidents have at least one factor in common: the human factor. The actions, decisions, thoughts, or perceptions of one or more humans contributed to the accident. These are just a few examples of safety problems researched by the Human Factors Group of the Pittsburgh Research Center, U.S. Department of Energy.

In the October 1998 issue of the Holmes Safety Association Bulletin, NIOSH authors Mark Stephenson and Carol Merry wrote the article Hearing loss among miners and measures to protect hearing, which focused on the extensiveness of hearing loss among miners and the use of hearing protection to prevent hearing loss. While the primary importance of engineering controls in preventing hearing loss was discussed, since miners do not generally have control over their work en­vironment or assignments, the article focused on the importance of consis­tent use of hearing protection. In this article, all aspects of a compre­hensive approach to the prevention of hearing loss in miners will be covered. The discussions will do the following: ? describe application of the hierarchy of controls in preventing hear­ing loss in miners, ? give an example for each type of control measure and its impact, ? explain the major provisions, be­yond the hierarchy of controls, of the recent NIOSH document en-titled Criteria for a Recommended Standard: Occupa­tional Noise Exposure, Revised Criteria 1998 relative to a hearing loss prevention program, and ? summarize important points on control of noise exposure for every miner to remember, equipment in most facets of operations. Miners are well aware of noisy areas in or around equipment, icluding extraction machinery such as draglines, power shovels, longwall shearers, and continuous mining machines; powered haulage equipment such as load-haul-dumps, shuttle cars, large trucks, and continuous haulage units; roof bolting machines; jackleg drills and stopers; auxiliary equipment such as compressors, fans, and pumps; size reduction equipment, etc. They are not well aware of the insidious nature of pro­gressive hearing loss, which is per­manent, but they are generally aware of what levels of noise are hazardous and what measures can be taken to reduce their personal noise exposure. Behaviorally, however, there are many reasons why miners choose not to use hearing protection faithfully.

The fibrogenic effects of silica in experimental animals and humans has been extensively studied (I 7, 20). However, the pathogenesis and molecular mechanisms underlying lung injury and fibrosis resulting from silica exposure remain to be elucidated. It is well-documented that silica can stimulate monocyte-macrophage lineage cells to produce cytokines and growth factors, including interleukin-I (IL- I) (20), tumor necrosis factor-ex (TNF-cx) (I 7), insulin-like growth factor-I (IGF-1) (4), and transforming growth factor-P (TGF-P) (24). Consequently, it has been suggested that the fibrogenic effect of silica may be mediated by cytokines in the fater stages of silicosis. During the early phase of interaction between silica and the pulmonary alveolar macrophage, several direct and indirect lines of evidence suggest that reactive oxygen intermediates (ROI) may play a pivotal role in silica- induced tissue damage (2I, 22). One of the most important components of and contributors to ROI is nitric oxide (NO) produced from L-arginine and molecular oxygen under the catalytic influence of NO synthase (NOS) (I 9). At present, at least three NOS genes have been cloned: a constitutive neuron NOS, a constitutive endothelial cell NOS and an inducible NOS (iNOS) (16). A number of factors have been shown to differentially induce iNOS gene expression in different cell lines including bacterial cell products and foreign antigens. During inflammation, NO is a double-edged sword in terms of the host immune defense mechanisms and the physiologic response to an inflammatory insult. The macrophage is one of the major cellular sources of NO. During the early phase of acute inflammation, NO produced from the macrophage appears to facilitate the clearance of pathogens and promotes a local relaxation effect on the microcirculation of vascular smooth muscle. When the production of NO is left unattenuated, especially under oxidative stress, direct cytotoxic effects of NO can emerge through the formation of peroxynitrite as a result of a reaction between NO and superoxide (2).

"Interest in the fine structure of soots and carbon blacks is motivated by a variety of possible applications. The structure provides information on the origins of the particles and on their adsorptive and reactive properties. This paper describes a method for quantification of the structure of soots and carbon blacks based on direct electron microscopic observation followed by image analysis of these materials. High-resolution transmission electron microscopy (HRTEM) provides a very detailed observation of particle structure. The differences in soot structure, because of its complexity, may not be easily quantifiable with the human eye; therefore, high-level computer software has been used to manipulate HRTEM images. This technique involves the application of fast Fourier transforms (FFT) to single particles and the measurement of characteristic parameters such as interplanar spacings and crystallite sizes from these particles. The methodology and application of this characterization technique are presented here. Results are shown for dif¬ferent samples obtained from soot and carbon black particles selected to illustrate the capabilities of the methodology. Quantitative information can be obtained on structural characteristics, e.g., interplanar spacing, circularity, orientation, elongation, and length distribution of lattice fringes, as well as on the fractional coverage of the extracted pattern. c 1996 Wiley-Liss, Inc.This paper presents a combination of techniques for recognizing and classifying different carbonaceous materials, using their structural morphology. The value of the electron microscope in studying the morphology of such materials has been well-established, and abundant information is available (Ishiguro et al., 1991; Lahaye and Prado, 1978; Buseck, 1992).The use of HRTEM has brought further insight into this matter. Qualitative assessment of the structural order of such materials has been attempted over a long period of time in order to differentiate between various sources or treatments. There is, to date, no successful method available for quantifying the structural characteristics of various carbonaceous materials. This paper presents a technique which is able to recognize dif-ferent materials and to quantify the structural parameters of those materials using HRTEM and computational image analysis, including pattern recognition techniques.In the case of perfectly ordered graphitic crystals, electron diffraction is widely used for obtaining details on structure. Analysis of diffraction patterns can reveal parameters such as interplanar spacing, d002, crystallite size along the c-axis, Lc, crystal size in the plane of the layers, L., mean number of crystallites, Nc, etc.Complicated electron microscopy methods such as defocusing, tilted illumination, axial illumination, etc., often have to be used in order to characterize complex materials such as randomly oriented crystals in soots and carbons. Round Mountain Gold (RMG) in central Nevada, reports that over a nine-year period and a million operating hours, its fleet of 150-ton and 190-ton haul trucks has averaged better than 93% availability, with some of the trucks approaching 70,000 operating hours. Repair costs have been far below industry averages. Mine operations manager Pete Kesl credits the high availability and low repair costs to a succession of Product Support Agreements between RMG and Caterpillar dealer Cashman Equipment. He adds that good equipment and a high level of trust and co­operation between all parties to these agreements have been essential to their success at Round Mountain.

Humankind&apos;s interest in measurement of human physical strength probably dates to the first humans. At that time, life was truly a struggle in which the fittest survived. To a great extent, fittest meant strongest. Interestingly, current interest in human physical strength in the workplace stems from 1970-1980s vintage research demonstrating that persons with adequate physical strength are less likely to be injured on physically demanding jobs. Survival in many modem workplaces may still be a case of survival of the strongest. There is, however, a flip side to this issue - that persons with limited strength are more likely to be injured on "hard" jobs. To address this problem, we can apply what we know about physical strength to job design. "Hard" jobs can be redesigned to be within the physical strength capability of most people. Since physical strength is important to these jobs, we must find ways to quantify it through testing. This publication concerns human physical strength testing. Its purpose is not to recommend any particular type of testing, but rather to describe the types of testing available and their uses. It is up to each individual user of strength testing to decide which testing technique is most appropriate for his or her particular application. This booklet discusses four types of strength testing: isometric, isoinertial, psychophysical, and isokinetic. Human Strength Before describing the different types of strength measurement, we must define the term "strength" and explain the concept of strength measurement. Strength is defined as the capacity to produce force or torque with voluntary muscle contraction. Maximum strength is defined as the capacity to produce force or torque with a maximum voluntary muscle contraction.(1,2) These definitions include some key words that must be explained. A voluntary muscle contraction is "voluntary." When a person&apos;s physical strength is measured, only the effort the person willingly puts forth at the time is measured. Thus, when we test a person&apos;s "maximum strength," we are not measuring his or her actual maximum, but some lesser value representing what he or she is comfortable expressing at the time with the existing equipment and environmental conditions. Interestingly, when researchers startled persons being tested (e.g., by setting off a starter&apos;s pistol behind them), they have found significant increases in measured strength.(3) It has been hypothesized that the lower strength displayed during normal testing provides a margin of safety against overloading and damaging muscle tissue. The test equipment and the tested person&apos;s familiarity with the process also influence the "voluntary" strength output. The interface between the tested person and the test equipment is particularly important. A poorly designed interface induces localized tissue pressures that vary from uncomfortable to painful. In this situation, testers are measuring voluntary discomfort tolerance - not strength. It is important for strength researchers to keep the "voluntary" nature of their data in mind when they are designing their equipment and protocols.

"I. INTRODUCTION(SILICA, THE LUNG, INFLAMMATION, AND FIBROSIS)Inhalation of respirable-size silica (0. I to 5 µm in diameter) deposits the particles in the deep spaces of the Jung where oxygen is exchanged. The resulting pattern of inflammation is substantially different compared to that observed from other inhaled substances such as carbon, polystyrene titanium dioxide etc. 1 Following the inflammatory reaction the body attempts to isolate the damage by walling off the area via fibrosis. 2 The unique nature of the inflammatory response to silica makes it logical to infer that fibrosis is a later-stage manifestation of the initial inflammatory damage. In addition to the inflammation extensive lysis and cellular death particularly to macrophages and neutrophils, can occur, depending on the dose of silica.' The ingestion of significant amounts of silica by macrophages and neutrophils produces an inflammatory sequence which activates the lipoxygenase pathway of arachidonic acid metabolism and produces platelet activating factor (PAF) and leukotrienes (LT).4·5II. CALCIUM IONOPHORE MIMICS SILICA TOXICITY IN VITROPhagocytic cells exposed to silica in vitro produce PAF and LT. This response is reminiscent of phagocytic cells exposed to the calcium ionophore (A23 l 87) in the presence of ionic calcium.6-8 A23 l 87 forms a complex with free ionic calcium which passes through cellular membranes, thus transporting calcium inside the cell. The increased intracellular calcium concentration triggers the production of small amounts of prostaglandins large amounts of LT. PAF and. depending on dose cellular death (see Figure 1 ). Could silica cause LT production and cellular death by a similar mechanism? How could silica - an inert solid - act in a similar fashion as a soluble stimulant? Clearly the negative SiO2 and OH- surface of silica can bind calcium ions9 and therefore could theoretically act as a solid-state ionophore releasing bound calcium inside of cells following phagocytosis."

As technology evolves, accidents may occur because human-system interactions were not considered adequately in the process. A systematic methodology can be used to evaluate the causes of mishaps and to develop recommendations that will enhance safety. A recent trend observed in underground coal mining is used to illustrate this principle because an underground mine is a dynamic work environment. Mining is characterized not only by frequent geologic changes but by technological evolution that can seriously degrade human performance and compromise worker health and safety The case exemplified in this paper involves implementation of remotely controlled equipment for extended cut mining. Extended cut mining technology evolved with minimal ergonomic consideration even though the change from on¬board to remote machine operation dramatically modified the role of the human component. In response to concern that system interactions should be examined more closely. a methodology was developed to identify hazards.

Physiol. 14): L215-L223, 1996.Previous investigations demonstrated that type II pulmonary epithelial cells regulate extracellular matrix deposition as a function of time in primary culture. In those studies, the matrix fraction was analyzed as a whole. The present work focused on two components of the type II cell matrix, fibronectin and laminin. These glycoproteins have differing effects on differentiation of type II cells in primary culture. Fibronectin synthesis was quantitated between day 1 and day 6 in the cells, matrix, and medium; laminin synthesis was quantitated only in the cells. Although total fibronectin synthesis was regulated as a function of time in culture, reaching its greatest value on day 2, the average proportion of newly synthesized fibronectin in the cells (35%), medium (50%), and matrix (15%) remained constant over a 6-day interval. Between day 2 and day 6, the relative abundance of fibronectin messenger RNA increased 6.5-fold. Rates of cellular laminin synthesis did not vary with time in culture. These results support a role for differential regulation of fibronectin and laminin synthesis to determine the composition of the type II cell extracellular matrix. INTERACTIONS BETWEEN THE pulmonary epithelium and alveolar basement membrane may play a role in the physiology and pathophysiology of the lung. Pulmonary epithelial cells and the alveolar basement membrane form one component of the blood-gas diffusion barrier. Integrity of this barrier is crucial to normal lung function. Specific domains of alveolar basement membrane components have been proposed to determine localization of type I and type II pulmonary epithelial cells during lung growth and development, as well as during repair of the alveolar surface after injury (13). In the injured lung, the extent of damage to the epithelium and the underlying basement membrane is thought to be a key factor to determine subsequent repair or fibrotic scarring (3)."
